Improvement of desulfurizing activity of haloalkaliphilic Thialkalivibrio versutus SOB306 with the expression of Vitreoscilla hemoglobin gene

Abstract: A conjugation transfer and an expression system for Thialkalivibrio, has been developed for the first time and used for expression of VHb to improve desulfurizing activity.

“…In terms of the development of bacteria for desulfurization by metabolic engineering, expression of Vitreoscilla globin gene ( vgb ) and knockout of sulfate producing key gene hdrB improved desulfurizing activity using haloalkaliphilic T . versutus (Mu et al 2017 ; Sharshar et al 2020 ). Fed-batch culture coupled with design, build, test, and validate approach successfully led to the first tight inducible system construction in T .…”

“…The members of this genus are well adapted to hypersaline (up to salt saturation) and highly alkaline (up to pH 10.5) conditions and fix inorganic carbon as a carbon source (Sorokin et al 2006 ). It has been established that the Vitreoscilla hemoglobin (VHb) level in Thioalkalivibrio versutus D301 promoted thiosulfate oxidation (Mu et al 2017 ). The production of nanometric sulfur from sulfide was enhanced by 166.7 % in another T. versutus D301 mutant in which the conversion of sulfur to sulfate was blocked by deleting the critical hdrB gene (Sharshar et al 2020 ).…”

“…The native vgb promoter works in several Gram-negative bacteria, including eight-tandem vgb promoter P8vgb in E. coli , Halomonas bluephagenesis and Halomonas campaniensis [ 37 , 38 ]. The specific promoters that have been chosen for other bacteria include trc promoter in E. coli [ 39 , 40 ], tac promoter in E. coli and Thialkalivibrio versutus [ 41 , 42 , 43 ], P phaC1-j5 promoter in Cupriavidus necator [ 44 ], and P43 promoter in Bacillus subtilis [ 45 ]. Fungal promoters that have been used for expression in fungi include: tubulin promoter in Aurantiochytrium sp.…”

“…Dependent on the function of sulfide receptor and storage [ 23 ], the expression of VHb in Rhodococcus erythropolis (desulfurization bacterium) presented a higher desulfurization ratio than the control ( vgb + -37.5% and vgb − -20.5%) under hypoxic conditions [ 83 ]. In the following, an increase of 11.7 ± 1.8% on the rate of thiosulfate scavenge was achieved in T. versutus by introducing the vgb gene [ 42 ]. In addition, the co-culture of desulfurization bacteria is also an effective sulfur degradation method [ 84 ].…”

“…established a highly efficient genetic transformation system based on the conjugative transfer method in the genus Thialkalivibrio . Meanwhile, the average thiosulfate removal rate of T. versutus SOB306 was improved 11.7 ± 1.8% by expressing Vitreoscilla hemoglobin (VHb), because it utilizes dissolved O 2 more efficiently with the help of VHb [68]. With further research, Sharshar et al.…”
