Improved technique for the expression of fragile-X in cultured amniotic fluid cells

Abstract: An improved technique for inducing fra(X) expression in cultured cells was obtained by using diazepam for mitotic arrest and 5-fluorodeoxyuridine (FUdR) for the induction of fra(X) expression. The method was developed using cultured fibroblast and urinary cells from fra(X) patients. Prenatal studies were performed on cultured amniotic fluid cells in five pregnancies at risk for fra(X). In two cases the cultured cells showed a 46,XY, fra(X) karyotype. One of the pregnancies was terminated and the diagnosis was … Show more

“…Prenatal diagnosis was performed on 13 pregnancies at risk for fra(X). There have been other reports in the literature concerning prenatal diagnosis of fra(X) (Shapiro and Wilmot, 1986;Shapiro et al, 1988;Purvis Smith et al, 1988;Tommerup et al, 1986;McKinley et al, 1988;Jenkins et al, 1988;Tejada et al, 1983;Von Koskull et al, 1985;Schmidt and Passarge, 1986). Most of these studies present data obtained from methods using different culture media or induction treatments.…”

“…The fragile X (fra(X)) syndrome is one of the major causes of mental retardation among mentally retarded boys next to trisomy 21 (Von Koskull et al, 1985). Since 1980, we have had the opportunity to study the expression of fra(X) in 30 families.…”
