Improved serodiagnosis of alveolar echinococcosis of humans using anin vitro-producedEchinococcus multilocularisantigen

Abstract: Serology is an important tool for the diagnosis of alveolar echinococcosis (AE) in humans. In order to improve serodiagnostic performance, we have developed an in vitro-produced Echinococcus mulilocularis metacestode vesicle fluid (EmVF) antigen for application in an immunoblot assay. Immunoblot analysis of EmVF revealed an abundant immunoreactive band triplet of 20-22 kDa, achieving a sensitivity of 100 % based on the testing of sera from 62 pre-operative and pre-treatment cases of active and inactive AE. Thu… Show more

“…For preparation of E. multilocularis vesicular fluid, metacestode tissue was cultivated in vitro, resulting in the formation of metacestode vesicles, as described by Müller and colleagues [23]. Fertile E. granulosus cysts were collected from sheep in Kyrgyzstan.…”

“…When the antigenic properties of EmHF cultured in vitro were investigated, some contradictions were observed. There was low antigenicity of EmHF obtained from the in vitro culture, and there was no reaction for low-molecular-mass proteins of Ͻ15 kDa (27). In contrast, an immunoblotting study revealed relatively strong antibody responses to Ͻ20-kDa proteins, although whether or not EmAgB3 was in those fractions was not determined (29).…”

“…Antibody responses became stronger over time. Several proteins, including those corresponding to molecular masses of 21 and 23 kDa, which showed prominent reactions (27) exhibited antibody responses 3 months postinfection. However, these proteins were cross-reactive with CE sera (Fig.…”

“…Western Blot assays are already used for diagnostic investigations in other parasitic diseases like echinococcosis and cysticercosis in humans (Gekeler et al 2002;Muller et al 2007) or infections in cattle with Neospora caninum and Besnoitia besnoiti (Cortes et al 2006). For serological testing in humans for Trichinella spp., commercialized Western Blot assays are routinely used as a confirmatory test to distinguish between patients with Trichinella infections and other helminth infections (Robert et al 1996;Kociecka 2000;Yera et al 2003).…”
