Improved sensitivity for detection of breast cancer by combination of miR-34a and tumor markers CA 15-3 or CEA

Abstract: BackgroundMicroRNAs biomarkers have shown value for diagnosis and prognosis of various cancers. Combination with established tumor markers has rarely been done.ResultsBreast cancer patients had significantly higher serum RNA loads (AUC 0.665), lower miR-34a (AUC 0.772), higher CEA and CA 15-3 levels (AUCs 0.717 and 0.721) than healthy controls. miR-34a correlated with tumor stage and hormone receptor status. There was no significant difference between groups for all other miRNAs. Combination of miR-34a with CE… Show more

“…Currently, 30424 mature microRNAs have been reported and more than 2500 mature miRNAs have been discovered in mammalian systems [7]. Many studies observed elevated levels of miRNAs in the circulation from breast cancer patients, such as miR-21 [8], miR-20b-5p [9], miR-34a [10] [11] [12], miR-133a [13] [14], miR-145 [15], miR-181a [16], miR-424 [17], miR-801 [18] [19], and so on [20] [26]. According to the GEN-CODE analysis 27817 transcripts originating from 15931 genes can be identified as lncRNAs [27].…”

“…In humans, over 1000 miRNAs have been identified, and each miRNA potentially represses hundreds of target mRNAs [3]. Over the last several decades, there have been numerous studies that have found associations between miRNAs and various types of cancer, such as lymphocytic leukemia [4,5], lung cancer [6,7], colorectal neoplasia [8], Burkitt lymphoma [9], glioblastoma [10], tumor cell [11], B cell lymphomas of hepatocellular carcinoma (HCC) [12], oral squamous cell carcinoma [13], and breast cancer [14]. Relatedly, many biological techniques have been developed for miRNA detectors, including Northern blot [15], real-time PCR [16], oligonucleotide microarray [17,18], laser-induced fluorescence [19], microarray [20,21], and even various label-free techniques [22], as well as some strategy approaches for the detection of miRNAs, such as electrochemical, photoelectrochemical and optical fiber sensing methods.…”
