Increases in food production and the ever-present threat of food contamination from microbiological and chemical sources have led the food industry and regulators to pursue rapid, inexpensive methods of analysis to safeguard the health and safety of the consumer. Although sophisticated techniques such as chromatography and spectrometry provide more accurate and conclusive results, screening tests allow a much higher throughput of samples at a lower cost and with less operator training, so larger numbers of samples can be analysed. Biosensors combine a biological recognition element (enzyme, antibody, receptor) with a transducer to produce a measurable signal proportional to the extent of interaction between the recognition element and the analyte. The different uses of the biosensing instrumentation available today are extremely varied, with food analysis as an emerging and growing application. The advantages offered by biosensors over other screening methods such as radioimmunoassay, enzyme-linked immunosorbent assay, fluorescence immunoassay and luminescence immunoassay, with respect to food analysis, include automation, improved reproducibility, speed of analysis and real-time analysis. This article will provide a brief footing in history before reviewing the latest developments in biosensor applications for analysis of food contaminants (January 2007 to December 2010), focusing on the detection of pathogens, toxins, pesticides and veterinary drug residues by biosensors, with emphasis on articles showing data in food matrices. The main areas of development common to these groups of contaminants include multiplexing, the ability to simultaneously analyse a sample for more than one contaminant and portability. Biosensors currently have an important role in food safety; further advances in the technology, reagents and sample handling will surely reinforce this position.

A multiresidue method for the determination of seven nitroimidazoles and their hydroxy metabolites in milk was developed. Milk samples were extracted with acetonitrile and cleaned up on strong cation-exchange solid phase extraction cartridges. Evaporated to dryness first, the extracts obtained were then reconstituted in 0.1% formic acid and injected onto a liquid chromatography-tandem mass spectrometer (LC-MS/MS). The separation of analytes was achieved on gradient mode using a C18 column and a mobile phase consisting of 0.1% formic acid in acetonitrile and 0.1% formic acid in water. Multiple reaction monitoring mode was set for the MS/MS with positive electrospray ionisation. For quantification, the isotope dilution method was used with isotopically labeled analogues of the target analytes. The method was evaluated entirely in accordance with EU Commission Decision 2002/657/EC and all validation criteria were in the required ranges. The method can easily detect and confirm metronidazole, dimetridazole, ronidazole, ipronidazole, and their hydroxy metabolites below the recommended concentration level of 3 µg/kg. The decision limits and detection capabilities ranged from 0.11 µg/kg to 0.22 µg/kg and from 0.19 µg/kg to 0.37 µg/kg respectively. The overall recoveries were between 96.6% and 105.2% with a good coefficient of variation, less than 8.7% under within-laboratory reproducibility conditions.

We present a high-throughput confirmatory method for the analysis of 11 nitroimidazoles in muscle samples (metronidazole, MNZ; dimetridazole, DMZ; ronidazole, RNZ; tinidazole, TNZ; carnidazole, CRZ; secnidazole semihydrate, SCZ; ornidazole, ORZ; metronidazole-hydroxy, MNZ-OH; ipronidazole, IPZ; ipronidazole-hydroxy, IPZ-OH; and dimetridazole-hydroxy, HMMNI). Extraction with acetonitrile is efficient and simple, with the majority of recoveries ranging between 65 and 101%, and without the need for clean-up of the extracts. The chromatographic analysis is performed using UPLC-QqQ-MS in the MRM mode, with an electrospray source operated in positive mode. Total chromatographic analysis time is 12 min. This method has been fully validated in muscle according to Decision 657/2002/CE guidelines, and the CCα values obtained were in the range of 0.04-0.4 μg·kg(-1). The method is currently used for the analysis of muscle samples and has been tested in other matrices of animal origin, such as kidney, retina and egg, with excellent results.
