Improved media for normal human muscle satellite cells: Serum-free clonal growth and enhanced growth with low serum

Abstract: We have developed a serum-free medium for clonal growth of normal human muscle satellite cells (HMSC). It consists of an optimized nutrient medium, MCDB 120, plus a serum-free supplement, designated SF, that contains epidermal growth factor (EGF), insulin, dexamethasone, bovine serum albumin, and fetuin. Fibroblast growth factor was needed with dialyzed fetal bovine serum (dFBS) as the only other supplement, but in media containing SF, it was only slightly beneficial, and was omitted from the final medium with… Show more

“…RT-PCR-5 g of total cellular RNA was reverse transcribed using 100 units of Superscript reverse transcriptase (Life Technologies, Inc., Eggenstein, Germany) and oligo(dT) [12][13][14][15][16][17] as a primer. Human ␣NAC and skNAC fragments were amplified by PCR.…”

“…The skeletal muscle growth medium (GM) is a modification of that of Ham et al (1988): MCDB-131 with -glutamine, 50 IU/ml penicillin, 5 µg/ml streptomycin, 2·5 µg/ml amphotericin B, 0·4 µg/ml dexamethasone, 0·5 mg/ml bovine serum albumin (BSA), 0·5 mg/ml fetuin, and 10% fetal calf serum (FCS) (GIBCO, Paisley, UK). Initially, 50 µg/ml insulin (Novo Nordisk, Crawley, West Sussex, UK) was included in the GM, as suggested by Ham et al (1988); we subsequently found that cells grew equally well in the absence of insulin. Skeletal muscle fusion medium (FM) consisted of minimum essential medium (MEM)-alpha modification with 50 IU/ml penicillin, 5 µg/ml streptomycin, 1% -glutamine and 2% FCS (GIBCO).…”
