Abstract

An improved HPLC assay with ultraviolet detection for the simultaneous determination of cortisol and cortisone in urine is described. In contrast to previously reported sample extraction procedures, here a solvent extraction with diehloromethane is performed first followed by a solid-phase extraction using CIS-extraction cartridges. The method that applies a gradient elution system with methanol, acetonitrile, and water proved linear (30-3000 nmol/1), reproducible (CV 8.0-9.2%), and specific.Zusammenfassung: Es wird eine verbesserte HPLC-Methode mit UV-Detektion für die simultane Bestimmung von Cortisol und Cortison im Urin beschrieben. Im Gegensatz zu bisher beschriebenen Probenextraktionsverfahren wird bei dieser Methode zuerst eine Flüssigphasenextraktion mit Dichlormethan durchgeführt, der anschließend eine Festphasenextraktion mittels einer C18-Extraktionskartusche folgt.
