Improved Core Viscosity Achieved by PDLLA10kCo-Incorporation Promoted Drug Loading and Stability of mPEG2k-b-PDLLA2.4k Micelles

Abstract: Purpose This study aims to investigate the effect of poly(D, Llactic acid) 10K (PDLLA 10K ) incorporation on the drug loading and stability of poly(ethylene glycol) 2K -block-poly(D, Llactide) 2.4K (mPEG 2k -b-PDLLA 2.4k ) micelles. In addition, a suitable lyophilization protector was screened for this micelle to obtain favorable lyophilized products. Methods The incorporation ratios of PDLLA 10k were screened based on the particle size and drug loading. The dynamic stability, core viscosity, drug release, sta… Show more

“…Similar trends were also noted for the PEG 10 PCL y series (SI, Figure S10), indicating that even prior to lyophilization/reconstitution, the addition of β-CD promoted significant aggregation and morphological changes in the micelles when the PEG M n was >2 kDa. This behavior may be related to the formation of pseudo-rotaxanes between the β-CD and the PEG corona of the micelles, with subsequent hydrogen-bonding between the β-CD leading to aggregation of the micelles [46][47][48][49]. β-CD is also known to form inclusion complexes with PCL, which may also contribute to the disruption of the micelle morphology [50][51][52][53].…”

“…ter, with normal micelles being the carriers of choice, while hydrophilic drugs can be delivered using reversed micelles. 99 This makes them ideal for targeting ocular tissue and increasing corneal permeability when applied topically. Micelles, despite their challenges in drug loading, scalability, and potential toxicity from surfactant use, offer notable benefits for DED treatment.…”
