Improved Colorimetric Determination of Cell Wall Chitin in Wood Decay Fungi

Abstract: The Svennerholm modification of the Elson-Morgan method for glucosamine analysis was evaluated for its applicability to the rapid determination of chitin in wood decay fungi. The evaluation included extent of chromogen interference, sensitivity, color stability, and hydrolysis conditions for maximum release of glucosamine from fungal cell walls. With our further modification, the Svennerholm method was shown to be suitable for rapid quantitative determination of fungal chitin without chromatographic separation… Show more

“…trimethylsilyl derivatives by GLC-MS. Infrared (IR) spectra are obtained using the KBr technique (Price, 1972). Aminosugar content is determined after hydrolysis of the polysaccharides according to Chen & Johnson (1983), and these compounds are identified in an amino acid analyser. The absolute configuration of the monosaccharides is determined after hydrolysis of the polysaccharides and derivatization according to Gerwig et al (1979) by GLC-MS of the tetra-O-TMSi-(1)-2-butylglycosides obtained.…”

“…Total neutral polysaccharides were analyzed by anthrone reagent with glucose as standard (Fairbairn 1953), and chitin was measured colorimetrically with 3-methyl-2-benzothiazolone hydrazone by the Ride and Drysdale (1972) method. Total and extracted chitin in AIS was determined after acid hydrolysis as glucosamine (Chen and Johnson 1983).…”

“…In AIS and in all fractions, total neutral carbohydrates were analyzed by anthrone reagent with glucose as standard (Fairbairn 1953), and chitin was measured by the Ride and Drysdale (1972) method. Total chitin in AIS was determined after acid hydrolysis (Chen and Johnson 1983). Monosaccharides in hydrolyzed alcohol insoluble solids were analyzed by GC as alditol acetates (Blakeney and others 1983).…”
