Improved Brain Delivery of Pueraria Flavones Via Intranasal Administration Of Borneol-Modified Solid Lipid Nanoparticles

Abstract: Aim: To improve the drug delivery to the brain with borneol (Bo)-modified solid lipid nanoparticles (SLNs) of pueraria flavones (PTF) via intranasal administration. Materials & methods: PTF-loaded SLNs were modified with Bo by physical and chemical methods to synthesize PTF-Bo-SA-SLNs and PTF-Bo-SLNs. The prepared SLNs were characterized and their brain delivery effects were evaluated in vitro and in vivo. Results: There was a more pronounced accumulation of PTF-Bo-SA-SLNs in Caco-2 cells. Following intran… Show more

“…Since endocytosis of NPs by the BBB endothelial cells seems to be predominantly mediated by receptors, many efforts have been made with SLN surface functionalization to enhance their CNS availability (Ceña and Játiva, 2018;Kuo et al, 2019;Wang et al, 2019), as discussed in Section 2 for the functionalized angiopep-2 NPs to treat glioblastoma multiforme (Kadari et al, 2018).…”

“…Wang et al [ 75 ] studied the in vitro efficacy of intranasal Pueraria flavone solution, Pueraria flavone-loaded SLN, Pueraria flavone-loaded SLN modified with borneol and Pueraria flavone-loaded SLN modified with borneol and stearic acid. Pueraria flavone is extracted from the Pueraria thoom sonii and Pueraria lobata and is used for the management of CNS diseases, such as Parkinson’s disease and Alzheimer’s disease.…”

“…There are two main direct pathways of entry into the brain after intranasal administration, namely, the olfactory and the trigeminal pathway. PTF-Bo-SA-SLNs and PTF-Bo-SLNs can be transported directly from the nasal cavity into the brain through the olfactory region of the nasal epithelium and the trigeminal neural region avoiding the BBB ( 60 ). Furthermore, borneol co-modified liposomes loaded with palmitate azidothymidine have been shown to enhance the transportation of drugs to the brain and increase the accumulation of azidothymidine by 3.73-fold, indicating that the addition of 10% borneol enhances the transportation of drugs in the brain by loosening the tight junctions of BBB ( 61 ).…”
