Importance of particulate antigen for the induction of dual bronchial reaction in guinea-pigs

Abstract: There has been a lack of small animal models for the secondary allergic response (SAR) seen after bronchial challenge in many asthmatic patients. We have found that challenge with particulate instead of soluble antigen will provoke an SAR-like bronchial obstruction in the guinea-pig. The particulate form was obtained by coupling the antigen covalently to Sepharose beads (approximately 100 microns). Different experiments suggest that SAR is obtained only when the challenge is induced via IgE-mediated mechanisms… Show more

“…Recently we described the effect of budesonide (BUD), a new GCS, on an IgE-mediated bronchial anaphylactic reaction in sensitized guinea pigs [8,9]. The present investigation shows that BUD inhibits the allergen-in duced early and late reaction in a recently developed guinea pig model [ 10],…”

“…Recently a new model of antigen-induced late al lergic reaction has been developed [10], Briefly the model is characterized as follows. Instillation of a particulate antigen (OA-Sepharose) to guinea pigs sensitized to produce IgE antibodies results in an im mediate anaphylactic bronchial obstruction which declines within 90 min.…”

“…Antigen immobilization might be one of the facilitating factors for the induction of LPR. Wieslander et al 15 induced secondary allergic response to OA in guinea pigs by coupling the antigen covalently to Sepharose beads. Similarly, adjuvants such as alumen or Freund's adjuvant also retain antigen within the injection point.…”

“…Several guinea-pig models of asthma have been developed focusing on both the early and the late phase responses, and on the airways hyperresponsiveness induced either by antigen or environmental and occupational chemicals. Although anaesthetized guinea-pigs have been used to study these phenomena [5][6][7][8], models closer to human asthma have been developed by prolonged measurement of bronchial function in conscious animals using whole body plethysmography [9,10]. However, important methodological differences exist between individual models, in particular with the routes of allergen sensitization and challenge, i.e.…”
