Implementation of the plasma MYCN/NAGK ratio to detect MYCN amplification in patients with neuroblastoma

Abstract: Detection of amplification of the MYCN gene is essential for determining optimal treatment and estimating prognosis of patients with neuroblastoma (NB). DNA FISH with neuroblastoma tissues or patient-derived bone marrow cells is the standard clinical practice for the detection of MYCN amplification. As tumor cells may often be unavailable, we developed a method to detect MYCN amplification in the plasma of patients with neuroblastoma. Taking single-copy NAGK DNA as reference, we used real-time quantitative PCR… Show more

“…Our study considered the status of MYCN, 11q23 and 1p36 LOH. They were present in 16.2%, 30.5% and 33.1% of patients, respectively, which was consistent with previous reports 13,22 . Here, we found that only one patient presented with both MYCN amplification and 11q23 LOH.…”

“…Neuroblastoma (NB) is the most frequent extracranial solid tumor, which arises from neural crest cells of sympathetic nervous system 1 . It represents 8%–10% of all pediatric tumors and up to 15.84% in infants, but accounts for 10%–15% of cancer‐related deaths in children 2,3 .…”

“…Neuroblastoma (NB) is one of the most common malignant tumors in children, which originates from neural-crest cells, and related with about 12% to 15% of pediatric cancer deaths [ 1 , 2 ]. Approximately 50% of NB patients show extensive metastases at initial diagnosis, which would have a poor prognosis [ 3 , 4 ]. Bone marrow is the most common metastasis site of NB, and patients without bone marrow involvement (BMI) would have a more favorable prognosis than those with BMI [ 5 ].…”
