Impairment of social behaviors in Arhgef10 knockout mice

Abstract: BackgroundImpaired social interaction is one of the essential features of autism spectrum disorder (ASD). Our previous copy number variation (CNV) study discovered a novel deleted region associated with ASD. One of the genes included in the deleted region is ARHGEF10. A missense mutation of ARHGEF10 has been reported to be one of the contributing factors in several diseases of the central nervous system. However, the relationship between the loss of ARHGEF10 and the clinical symptoms of ASD is unclear.MethodsW… Show more

“…Social behaviors include social preference and social recognition (Albers, 2012;Tanimizu et al, 2017;Young, 2002). Social recognition consists of the behavioral characteristics of distinguishing a novel conspecific from a familiar one, which is evolutionarily well conserved because of its importance for survival (Bicks et al, 2015;Lu et al, 2018;Moy et al, 2004). Social recognition is impaired in several neurodevelopmental disorders, such as autism spectrum disorder (ASD) and schizophrenia (Carter et al, 2011;McKibben et al, 2014;Webb et al, 2010).…”

“…Neurotransmitters like 5-HT and DA are involved in the brain circuitry related to social behaviors (Miczek et al, 2002; Watanabe and Yamamoto, 2015; Lu et al, 2018). The dorsal striatum plays a role in internally guided social behavior, the ventral striatum regulates social behavior by the integration of external social stimuli (Báez-Mendoza and Schultz, 2013; van den Bos, 2015), and the balance of 5-HT and DA levels determines the role of each striatal region in social behaviors (van den Bos, 2015).…”

“…The Rho guanine nucleotide exchange factor 10 is highly expressed in the developing brain and thought to play a role in developmental myelination ( Verhoeven et al 2003 ). Arhgef10 knockout mice are a model for impaired social behaviors in autism ( Lu et al 2018 ). Moreover, ARHGEF10 has been associated with schizophrenia ( Jungerius et al 2008 ).…”
