Impact of wastewater effluent containing aged nanoparticles and other components on biological activities of the soil microbiome, Arabidopsis plants, and earthworms

Abstract: The amount of engineered nanomaterials (ENMs) in the environment has been increasing due to their industrial and commercial applications. Different types of metallic nanoparticles (NPs) have been detected in effluents from wastewater treatment plants (WWTPs). The effluents have been reclaimed for crop irrigation in many arid and semi-arid areas. Here, a soil micro-ecosystem was established including a microbiome, 4 Arabidopsis thaliana plants, and 3 Eisenia fetida earthworms, for a duration of 95 days. The imp… Show more

“…The widespread use of NP-containing products has led to the direct exposure of the terrestrial environment to these nanosized materials, raising concerns regarding their safety and biocompatibility with both living organisms and the environment [1,16,[98][99][100]. Therefore, the risks and hazard assessment of NP exposure for plants, soil organisms, and consequently, humans, as a result of contamination of the food chain need to be addressed [1,101,102].…”

“…Subsequently, a drop of the dispersion was applied to a holey carbon film and dried in oven at 60°C for 10 to 15 min for TEM observation. Size distribution of the nanoparticles was analyzed by dynamic light scattering according to our previous method (DLS, DynaPro NanoStar, Wyatt Technology, Santa Barbara, CA, USA) (Campobasso et al, 2020; Liu et al, 2018, 2019).…”

“…16S rRNA gene V4 variable‐region PCR primers 515/806 were used for PCR amplification. PCR and sequencing were performed according to our previously published method (Liu, Williams, et al, ). The PCR was performed under the following conditions: 94°C for 3 min; followed by 94°C for 30 s, 53°C for 40 s, and 72°C for 1 min for 28 cycles; and a final elongation step at 72°C for 5 min.…”
