Impact of UGT2B7 His268Tyr polymorphism on the outcome of adjuvant epirubicin treatment in breast cancer

Abstract: IntroductionEpirubicin is a common adjuvant treatment for breast cancer. It is mainly eliminated after glucuronidation through uridine diphosphate-glucuronosyltransferase 2B7 (UGT2B7). The present study aimed to describe the impact of the UGT2B7His268Tyr polymorphism on invasive disease-free survival in breast cancer patients after epirubicin treatment.MethodsThis is a pharmacogenetic study based on samples collected from 745 breast cancer patients of the Austrian Tumor of breast tissue: Incidence, Genetics, a… Show more

“…The impact of UGT2B7 polymorphisms on the metabolism of drugs used to treat breast cancer has also been investigated. For example, the UGT2B7 rs7668258 C>T polymorphism has been associated with the metabolism, efficacy and toxicity of epirubicin chemotherapy in breast cancer patients . Moreover, UGT2B7 802Tyr has been suggested as a candidate biomarker for a good response to tamoxifen therapy in patients with breast cancer …”

“…It also conjugates various classes of major drugs including analgesics, carboxylic non-steroidal anti-inflammatory drugs, and anti-carcinogens 9. In addition, UGT2B7 polymorphisms were found to have associations with an increased risk of bladder cancer, colorectal cancer, and a treatment method for breast cancer 10-1213.…”

“…UGT2B7 contributes to glucosidation of hydesoxycholic acid in the liver, as well as glucuronidation of steroid hormones and fatty acids, and to the detoxification of cholestatic bile acids and may have a role in the formation of proximal carcinogens, such as quinine estrogens . In patients with BC that carried the rs7439366, they might have had a benefit after adjuvant epirubicin‐based treatment . Nieuweboer et al .…”
