Impact of pre-existing dengue immunity on human antibody and memory B cell responses to Zika

Abstract: Little is known about enduring memory B cell (MBC) responses to Zika virus (ZIKV) and their relationship with circulating antibodies. Here we comprehensively assess MBC frequency and specificity alongside serum binding and neutralizing antibody responses to ZIKV ~2 weeks and ~8 months postinfection in 31 pediatric subjects with 0, 1 or >1 prior infections with the related dengue virus (DENV). ZIKV infection elicits a robust type-specific MBC response, and the majority of late convalescent anti-ZIKV serum neutr… Show more

“…A study showed that cross-reactive ZIKV-primed CD4 + T cells 73 . On the other hand, we showed that naïve animals with DENV de novo response did not cross-neutralized ZIKV at all, which state that although similar, antigenic differences are sufficient to mount predominantly type-specific rather than crossreactive responses during a primary infection 50,55 .…”

“…Mouse studies have shown that Bmem responses to West Nile virus (WNV) retain the ability to engage variant viruses whereas antibody derived from LLPC is poorly able to inhibit variant virus (Purtha et al, 2011). Studies of human Zika and Dengue virus responses have shown that Bmem responses are more diverse than those of the serum (LLPC)-derived antibody (Andrade et al, 2019).…”

“…Here, we described the development of an hAd5 ZIKV vaccine that encodes and expresses the full length prM and E structural proteins from ZIKV virus PRVABC59. Flavivirus E proteins are known to be the a target of neutralizing antibodies and the T cell response [13,40,41]. The goal for including prM in this vaccine construct is to aid in the proper folding of Zika E proteins to allow for the proper folding of E and the presentation of neutralizing antibody epitopes, leading to a strong antibody response that has been shown in other similar vaccine constructs [60,61].…”

“…The presence of these three structural proteins on the virus surface make them all possible antibody epitopes. Studies done to map the antibody response to ZIKV have shown that, similar to other flaviviruses, the bulk of the antibody response is directed toward the E protein [39][40][41]. Additionally, evidence from natural history studies indicate that ZIKV E stimulates both a T cell and antibody response in humans [42][43][44][45][46].…”
