Impact of lipopolysaccharides on cultivation and recombinant protein expression in human embryonal kidney (HEK‐293) cells

Abstract: The human embryonal kidney 293 cell (HEK‐293) is a widely used expression host for transient gene expression. The genes or plasmids used for the transient transfections are usually propagated and extracted from the gram‐negative bacterium Escherichia coli, the workhorse for molecular biologists. As a gram‐negative bacterium E. coli has an outer membrane (OM) containing lipopolysaccharides (LPS) or endotoxins. LPS are very potent inducers of inflammatory cytokines in the body. In early research phases DNA inten… Show more

“…It is worth noting that bactericidal activity was observed for P. aeruginosa ATCC 27853 at 32.50 μg ml −1 and P. aeruginosa P2307 at 65.00 μg ml −1 , which were the highly virulent strains against the G. mellonella infection model (). In the case of Gram-negative bacteria, the bacteriostatic effect can be desirable since the release of LPS in the body can be avoided [65, 66]. These results highlight the potential of this SAR-endolysin against a broad range of MDR and virulent Gram-negative bacteria.…”

“…In addition, DNA should be of high quality, purity, integrity and concentration [ 7 , 8 ]. For example, DNA preparation can be contaminated with endotoxins, which can alter the growth of some cell lines [ 8 , 9 ]. Lastly, the transfection method plays a critical role not only on the efficiency parameter but also on the cell survival.…”

“…The quality of DNA depends, among other factors, on the presence of endotoxins or lipopolysaccharides (LPS), which are byproducts of plasmid DNA preparation [ 7 ]. The presence of LPS is unlikely to significantly affect the transfection efficiency or cell proliferation of commonly used cell lines, but it may pose challenges for specific cell lines [ 7 , 8 , 9 ]. Nonetheless, the two major drawbacks of DNA-based transient transfection are the inability to carefully control the expression level of the foreign construct even by using weak versus strong promoters [ 10 ] and, for most cell lines, the inability to reach up to 95% of transfected cells [ 11 ].…”

“…A detailed analysis of different HEK293 cell lines can be found in the studies by Malm et al [ 19 ] and Tan et al [ 3 ]. HEK293 cells growing in suspension typically have mean cell diameters ranging from 14 to 16 [ 20 , 21 , 22 ] and a typical maximum specific growth rate ranging from 0.020 –0.029 [ 23 , 24 , 25 , 26 ]. Jang et al [ 24 ] were able to demonstrate comparable growth rates between adherent growing and in suspension growing HEK293 cells.…”
