Preventive effect of Stachys lavandulifolia against gentamicin-induced oxidative stress and nephrotoxicity in rats

Gentamicin (GM) is an effective antibiotic against gram-negative bacterial infection. However, GM can produce nephrotoxicity. Stachys lavandulifolia Vahl. (SL) is extensively used in folk medicine as herbal tea and has strong antioxidative effects. The aim of this study was to investigate the protective effect of SL on GM-induced nephrotoxicity in rats. In this study, forty-two male Wistar rats were randomly divided into six equal groups (N = 7 each): control (group I), GM-treated (group II), SL200 mg/kg + GM-treated (group III), SL400 mg/kg + GM-treated (group IV), SL200 mg/kg-treated (group V), and SL400 mg/kg-treated (group VI) groups. The treatment procedures continued for 10 days. At the end of the experiment, serum and the kidneys of all groups were harvested for histological and biochemical examination. No statistical significant improvement in histological and biochemical indices was shown at 200 mg/kg of SL treatment in group III compared with group II; however, at 400 mg/kg of SL, normalized serum levels of uric acid (P < 0.001), urea (P < 0.001), creatinine (P < 0.01), and total proteins (P < 0.05) were shown in group IV. Treatment with SL400 mg/kg also corrected the increased relative kidney weight in GM-treated animals (P < 0.05). GM nephrotoxicity was shown by elevated levels of malondialdehyde (P < 0.001) and decreased amounts of catalase (P < 0.001), superoxide dismutase (P < 0.01), and glutathione (P < 0.01) in renal tissue. The oxidative injury induced by GM was prevented by SL400 mg/kg treatment. Such treatment also normalized abnormal renal histology induced by GM. These data suggest that treatment with SL extract may be helpful to reduce gentamicin-induced nephrotoxicity.

Objectives: This study was delineated to investigate the ameliorative effect of Echinacea purpurea and curcumin against oxidative stress and immunosuppression induced by dexamethasone in rabbits. Methods: A total of 20 healthy rabbits were used in this study and were randomly classified into 4 groups of 5 each. The 1 st group (non-treated) was served as a control, the 2 nd group intramuscularly medicated with dexamethasone (2 mg/kg) 3 times at 6 hours interval, the 3 rd group medicated with dexamethasone at the same dose then received E. purpurea (130 mg/kg) orally day after day for 21 days, and the 4 th group medicated with dexamethasone at the same dose then received curcumin (400 mg/kg) orally every day for 21 days. Blood and serum samples were collected after 1, 7, and 14 days post-dosing. Results: Dexamethasone medicated group showed a significant decrease in WBCS, lymphocytes, neutrophils, and monocytes at the 1 st , 7 th, and 14 th day, but E. purpurea and curcumin medicated groups showed significant improvement in total and differential leucocytic count. Also, both plants showed a significant increase in lysozymes and nitric oxide against the suppressive effect of dexamethasone. Here, the dexamethasone group showed a significant decrease in serum catalase (CAT) and significant elevation in serum malondialdehyde (MDA) at 1 st and 7 th-day posttreatment. E. purpurea and curcumin groups showed a significant increase in serum CAT and significant depletion in MDA levels. Injection of dexamethasone significantly decreased the serum levels of total protein, albumin, and globulin as compared to the control group. These parameters were significantly increased after treatment with E. purpurea and curcumin. Treatment with E. purpurea or curcumin ameliorated tissue damages of liver, kidney, and spleen induced by dexamethasone. Conclusion: Treatment with Echinacea purpurea or curcumin could ameliorate oxidative stress and immunosuppression induced by dexamethasone in rabbits.

Three hundred Oreochromis niloticus were divided into five groups; the control fish (CNT) were fed a basal diet, beta glucan (βG) group was fed 0.5 g/kg−1 βG, atrazine (ATZ) group was exposed to 1/5 96‐hr LC50 (1.39 mg/L) ATZ, the (βG/ATZ) group was fed βG while being exposed to ATZ, and the (βG then ATZ) group was supplemented with βG for fifteen days before exposed to ATZ. ATZ exposure caused a decline in growth that was ameliorated by βG. ATZ reduced the levels of total and different types of leucocytes. Additionally, ATZ exposure caused reductions in total proteins, globulins, α 1‐globulin, α 2‐globulin, ɤ‐globulin, immunoglobulin M, lysozymes, superoxide dismutase, nitric oxide and catalase but increases in hepatic transaminases and malondialdehyde without any variations in albumin and β‐globulin. Exposure to ATZ also resulted in a rise in the mRNA level of IL‐8. In contrast, expression of IgM, SOD and CAT were decreased in the tilapias exposed to ATZ. Exposure to ATZ increases the susceptibility response to Aeromonas sobria challenge, as indicated by an increase in cumulative mortality post‐challenge. Supplementation with βG fifteen days before (βG then ATZ group), counteracted the adverse effects of ATZ on the immune, biochemical and antioxidants values, though only slight alleviation was observed with simultaneous treatment (βG/ATZ group). Our results established that ATZ has adverse impacts on immune responses, antioxidant equilibrium and its related genes. While, supplementation with βG before exposure to ATZ may be valuable for counteracting the possible damage caused by ATZ water pollution than its simultaneous treatment with ATZ.
