“…It has been shown by others that pCPA can block the pro-oestrous surges of prolactin and LH (Walker & Wilson, 1983;Horn & Fink, 1985) and also the surges of both hormones induced by administration of oestrogen or oestrogen plus progest¬ erone to chronically ovariectomized rats (Caligaris & Taleisnik, 1974;Coen & McKinnon, 1979;Iyengar & Rabii, 1983;Walker & Wilson, 1983;Meyer &Eadens, 1985). This effect of pCPA seems to be mediated by serotonin depletion or by the disappearance of the daily rhythm of serotonin (Walker & Wilson, 1983;Walker, 1984). The effect of pCPA on the surges of prolactin at pro-oestrus or in oestrogen-primed ovari¬ ectomized rats was also reversed by administration of 5-hydroxytryptophan, which restored serotonin syn¬ thesis (Caligaris & Taleisnik, 1974).…”

“…The lack of effects from dopaminergic ergot alkaloids on PVC in old rodents contrasts with widely cited observations that L-dopa reinitiates cycling from PVC in old rats. We note possible reasons for this difference: (1) differences between central effects of L-dopa and of Hydergine and bromocriptine (Vigouret et al, 1978); (2) decreased central dopaminergic activity may not be the primary cause for the onset of PVC (Forman et al, 1980;Walker, 1984); (3) the doses may have been suboptimal, as suggested by the L-dopa dose-response relationship (Linnoila and Cooper, 1976) and the varying effectiveness of L-dopa reported in different studies (Finch. 1979); and (4) the small ovarian reserve in acyclic, middle-aged C57BL/6J.…”

“…Thus, it has been suggested that simple depletion of these germinal centers accounts for reproductive senescence in women (7 Figure 2. Presumably, altered gonadotropin release results ultimately from CNS perturbations affecting at least the metabolism and secretion of monoamines (10)(11)(12). In general terms, these changes are reciprocal between catecholamines and serotonin (13), ultimately shifting the nature of monoamine signals to the pituitary from catecholamines to those predominated by serotonin.…”
