Cancer immunotherapy has been gradually introduced and has undergone noteworthy developments in recent years. The number of scientific publications has been expanding, and the progression in this field has been rapidly evolving with time. This study aimed to use bibliometric analysis to examine the past 20 years of research on cancer immunotherapy and identify future hotspots. A literature search for medical publications on immunotherapy in cancer from 2000 to 2021 was conducted in the Web of Science Core Collection on March 1, 2022. Visualization analysis was performed using VOSviewer software (version 1.6.16). From 2000 to 2021, a total of 18,778 publications were retrieved. Annual publication output grew rapidly from 366 in 2000 to 3,194 in 2021. The USA issued the largest number of publications ( n = 6,739, 35.89%), with the University of Texas System making the largest contribution ( n = 802, 4.27%). A total of 976 meaningful topics were identified and further classified into 4 different clusters (immune mechanism, cancer biology, immunotherapy and clinical trials). The most common research topics included ‘expression’, ‘chemotherapy’, ‘dendritic cells’, ‘pembrolizumab’ and ‘open-label’. Highly identified cancer types included hepatocellular, bladder, breast and lung cancer. A shift in popularity from mechanism research to clinical trials was observed, indicating that clinical application would be the center of attention in the future. Attention has been given to the field of cancer immunotherapy, and this trend will continue in the future. This study provides an unbiased visualization analysis on this topic in a scale-efficient manner for further research.

Top 100 most-cited articles on renal cell carcinoma: A bibliometric analysis

Background: To analyze the top 100 most-cited articles on renal cell carcinoma (RCC) using bibliometric methods based on the Web of Science core collection database and to explore the research status, hotspots, and emerging trends in RCC. Methods: The literature on RCC was searched in the Web of Science core collection database using a specific search strategy, and the types of literature were limited to articles and reviews, with no restrictions to language and publication date. The top 100 articles with the highest number of citations were extracted after the manual screening. The publication year, the number of citations, authors, country, institution, journal, and keywords of these articles were collected and analyzed. Descriptive statistics and visual analysis were performed using Microsoft Excel, VOSviewer, CiteSpace, R, and SPSS. Results: The number of citations of the top 100 articles varied from 541 to 4530, with a median citation count of 807.5, and the citation rates ranged from 13.8 to 448.4 citations per year. Motzer RJ (n = 22), Escudier B (n = 13), Rini BI (n = 13), and Hutson TE (n = 11) were major contributors to this research area, with Motzer RJ publishing 16 articles as the first author. The US (n = 73), France (n = 5), Canada (n = 4), and Sweden (n = 4) were the leading countries for RCC studies. MEMORIAL SLOAN KETTERING CANCER CENTER (n = 22) was the institution with the highest number of publications. These 100 articles were derived from 24 journals, and the New England Journal of Medicine had the largest number of articles published (n = 18, impact factor = 91.245). The keyword co-occurrence network analysis showed that research hotspots in this field included molecular mechanisms of RCC development and progression, surgical treatment, targeted drug-related clinical trials, and immunotherapy. Conclusion: We analyzed the top 100 articles with the highest number of citations in the field of RCC and identified the influential authors, countries, institutions, and journals in this field. This study also presented the current research status, hotspots, and future trends in RCC.

In 2020, bladder cancer, which commonly presents as urothelial carcinoma, became the 10th most common malignancy. For patients with metastatic urothelial carcinoma, the standard first-line treatment remains platinum-based chemotherapy, with immunotherapy serving as an alternative in cases of programmed death ligand 1 expression. However, treatment options become limited upon resistance to platinum and programmed death 1 or programmed death ligand 1 agents. Since the FDA’s approval of Enfortumab Vedotin and Sacituzumab Govitecan, the therapeutic landscape has expanded, heralding a shift towards antibody–drug conjugates as potential first-line therapies. Our review employed a robust scientometric approach to assess 475 publications on antibody–drug conjugates in urothelial carcinoma, revealing a surge in related studies since 2018, predominantly led by U.S. institutions. Moreover, 89 clinical trials were examined, with 36 in Phase II and 13 in Phase III, exploring antibody–drug conjugates as both monotherapies and in combination with other agents. Promisingly, novel targets like HER-2 and EpCAM exhibit substantial therapeutic potential. These findings affirm the increasing significance of antibody–drug conjugates in urothelial carcinoma treatment, transitioning them from posterior-line to frontline therapies. Future research is poised to focus on new therapeutic targets, combination therapy optimization, treatment personalization, exploration of double antibody-coupled drugs, and strategies to overcome drug resistance.
