Immunotargeted Photodynamic Therapy for Cholesteatoma

Abstract: Anti-EGFR ICG nanocapsules applied topically and activated as part of an IT-PDT scheme results in a high rate of cholesteatoma-derived keratinocyte cell death while negligibly affecting middle ear mucosal cells in vitro. These preliminary findings suggest that this is a feasible concept and that further investigation is warranted.

“…The nanocapsules were coated with an antibody against epidermal growth factor receptor (EGFR) to selectively target the keratinocytes. In vitro results showed selective targeting and killing of keratinocytes, but not mucosal cells after activation of the nanocapsules with infrared light ( Gluth et al, 2015 ).…”

“…The reported photosensitizers are mainly porphyrin-based derivatives ( Lucky et al, 2015 , Ormond and Freeman, 2013 ). Several classes of non-porphyrin derivatives, such as cyanines ( Fekrazad et al, 2015 , Gluth et al, 2015 ), xanthenes ( Gianotti et al, 2014 , Wang et al, 2013 , Yao et al, 2014 ), phenothiazines ( Samy et al, 2015 , Yu et al, 2015 ), and anthraquinones ( Sharma and Davids, 2012 , Theodossiou et al, 2009 ) were investigated recently. Most of the reported photosensitizers absorb light in the visible region.…”

“…Several methods, studied both in vivo and in vitro, have been proposed to inhibit or slow the growth of cholesteatoma (5)(6)(7)(8)(9)(10)(11). Residual cholesteatoma, caused by incomplete surgical removal of squamous epithelium from the middle ear, has been a well-known challenge (12)(13)(14).…”
