Immunosuppressive Compounds Affect the Fungal Growth and Viability of Defined Aspergillus Species

Abstract: Immunosuppressive drugs are administered to a number of patients; e.g., to allogeneic hematopoietic stem cell transplant recipients. Immunosuppressive drugs impair the immune system and thus increase the risk of invasive fungal disease, but may exhibit antifungal activity at the same time. We investigated the impact of various concentrations of three commonly used immunosuppressive compounds—cyclosporin A (CsA), methylprednisolone (mPRED), and mycophenolic acid (MPA)—on the growth and viability of five clinica… Show more

“…Interestingly, it is also reported to have antifungal activity especially on Aspergillus spp rendering it as a favorable therapeutic option. 69 Whereas the involvement of adaptive immunity 72 hours post-infection is accredited, the involvement of innate immunity is not inconsequential. Pathways like NOD-like receptor signaling pathway 70 and PI3K-Akt signaling pathways are reported to play a major role in immune cell activation especially neutrophils and mast cells suggesting that the innate immunity is still a significant part in the host-immune response.…”

“…Glucocorticosteroids cause wide-ranging immune aberrations increasing risk of IFRI in a dose dependent manner [16]. They also have direct species-specific effects promoting fungal growth [17]. Dexamethasone used in the context of coronavirus disease 2019 (COVID-19)-induced immunopathology and epithelial injury contributes to devastating fungal co-infections [18 ▪▪ ].…”

“…One of the most frequently used immunosuppressive mycophenolate mofetil, having established antifungal properties, can contribute to organ transplantation to eliminate commensal and unwanted fungal species or isolates. This immunosuppressive will consequently favor the invasive and mycophenolic acid-resistant species ( 379 ), hence could lead to invasive fungal infection burden, for example, in kidney recipients ( 380 ). After validation, this treatment direction can potentially lead to the development of molecular signatures of fungal resistance to immunosuppressants, which can be used as detecting tools to forecast clinical outcomes.…”
