Immunostaining of Pulpal Nerve Fibre Bundle/Arteriole Associations in Ground Serial Sections of Whole Human Teeth Embedded in Technovit® 9100

Abstract: A technique for embedding human undecalcified tooth specimens in Technovit® 9100 was developed, which permits immunohistological evaluation of pulp tissue in serial ground sections. Human molars were divided into 14-18 sections of about 23 µm thickness. Immunohistological double staining for S-100 and CD34 revealed unique associations of myelinated nerve fibre bundles with arterioles, which continued through the entire tooth pulp. These arterioles were not only accompanied by, but partially or totally envelope… Show more

“…Especially myelinated nerve bundles display a tight association with arterioles in the root pulp and in the central region of the crown pulp (Steiniger et al. ). Because neurites and blood vessels have been located in the same areas of the developing tooth in separate studies, this raises the question of their intimate association and developmental interdependence during development (Gaunt, ; Decker, ; Mohamed & Atkinson, ; Luukko, ; Kettunen et al.…”

“…Our method is associated with good preservation of different antigens in the sections. Other investigations have already demonstrated that embedding in Technovit ® 9100 is suitable for immunohistology [2628]. In general, we recommend using a high amplification method for immunostaining, such as a tyramide-enhanced immunoperoxidase procedure.…”

“…One gram of dibenzoylperoxide (hardener 1) was added to 200 ml of stabilized and destabilized base solution for the pre-infiltration 2 and pre-infiltration 3 solutions, respectively. Infiltration solution and stock solution A for polymerization (Table 1) were prepared in two steps as described by Steiniger et al (2013). For the infiltra-Q6 tion solution, 20 mg of polymethylmethacrylate powder (PMMA powder) was diluted in 160 ml of destabilized base solution.…”
