Immunoscintigraphy for ocular melanoma: a reliable diagnostic technique?

Abstract: We conclude that IS, using the antibody applied in this study, is of only limited value in patients with ocular melanoma. Our results suggest that antigenic differences, rather than histological characteristics or technical problems, are responsible for the low sensitivity in ocular melanoma compared to cutaneous melanoma.

However, their success has been limited because of the intrinsic limitations of radiolabeled antibodies and antibody fragments, such as slow circulation clearance ( , ) and reduced rates of tumor penetration ( , ).…”

“…Another class of promising agents for melanoma imaging and therapy are R-melanocyte stimulating hormone (R-MSH) peptide analogues. Wild-type R-MSH is a small tridecapeptide (Ac-Ser 1 -Tyr 2 -Ser 3 -Met 4 -Glu 5 -His 6 -Phe 7 -Arg 8 -Trp 9 -Gly 10 -Lys 11 -Pro 12 -Val 13 -NH 2 ), which is involved in the control of skin pigmentation. The biological activity of R-MSH is mediated through interactions with the melanocortin 1 (MC1) receptor (13).…”

“…The first immunohistological studies showed the binding of MoAb 225-28 S raised against cutaneous melanoma in choroidal melanoma (Bomanji et al 1987;Scheiffarth et al 1989). Many clinical studies were performed to assess the value of immunoscintigraphy (I S) in choroidal melanoma (Schaling et al 1989(Schaling et al , 1994Hungerford et al 1991;Stanowsky et al 1993;Lö ffler et al 1994) and the largest clinical trial was published on 101 cases (Scheidler et al 1992). It appears to be a valuable method in the diagnosis of choroidal melanoma and offers substantial sensitivity and specificity in the differentiation of intraocular lesions.…”
