Immunoradiometric Assay (IRMA) for Human Follicle Stimulating Hormone (FSH) and Luteinizing Hormone (LH) Using Common Avidin Solid Phase

Abstract: This paper describes the use of avidin-biotin interaction as an affinity system, wherein avidin immobilized magnetizable particles (cellulose) are used as a common separation system in immunoradiometric assay (IRMA) for hormones of the human reproductive system, human follicle stimulating hormone (FSH), and luteinizing hormone (LH). Biotinylated probe was prepared by biotinylation of specific monoclonal antibody for respective antigen using the caproyl derivative of biotin N-hydroxysuccinimide. The detector an… Show more

“…Several formats of IRMA employing range of antibodies and antibody fragments, variety of detection markers other than radioisotope and various solid support systems are available commercially [15,16] . The choice for the chosen system/format depends on the availability, affordability and existing infrastructural facilities with the manufacturer and users [17,18] .…”

“…radiolabeled antibody. Radiolabelled antibody at 1.5×10 5 cpm was generally used in our optimized assay design [8][9][10]15] . Increase in the labeled antibody corresponding to count rate from 1.0 to 3.0×10 5 cpm was studied in case of FSH IRMA, which led to encouraging results in estimated values for commercial QC samples.…”
