Immunophenotypic and immunogenotypic characteristics of TCRγδ+ T cell acute lymphoblastic leukemia

Abstract: Thirty T cell receptor (TCR)␥␦Firstly, more complete TCRD rearrangements were identified on the non-expressed allele in the non-V␦1 + group (83% vs 43%); secondly, a higher frequency of 'end-stage' J␥2.3 gene rearrangements was found in non-V␦1 cases on both TCRG alleles (83% vs 66%); thirdly, a higher frequency of complete TCRB rearrangements was found in non-V␦1 cases (79% vs 50%).

“…It should be noted that most TCRgd þ T-cell malignancies express Vd1 or another non-Vd2 gene segment in combination with a single Vg domain (generally not Vg9). 15,20 The detection of Igk-or Igl-restricted expression or monotypic Vb, Vg or Vd expression is relatively easy in flow cytometric studies of PB and bone marrow (BM) samples of patients with mature B-or T-cell leukemias. However, it appears to be more difficult in tissue samples with suspect lymphoproliferative disorders that are intermixed with normal (reactive) lymphocytes.…”

“…2 Incomplete and complete TCRB gene rearrangements were detected with TCRBD1U, TCRBD1, TCRBJ1, TCRBD2U, TCRBD2, TCRBJ2 and TCRBC probes (Ref. 47; DAKO Corporation, Carpinteria, CA, USA) in EcoRI and HindIII digests. TCRG gene rearrangements were analyzed with the J␥1.2 probe in BglII digests and the J␥1.3 and J␥2.1 probes in EcoRI digests.…”

“…This is most probably owing to the usage of the four different D␤ probes, which allow identification of incomplete V-D␤, D-D␤, and D-J␤ gene rearrangements. 47 Four other research groups analyzed and compared all three cross-lineage TCR genes in a total number of 100 cases. 5,7,10,11 Combining these published data and the data presented here show that TCRD gene rearrangements and/or deletions occurred in the majority of cases (ෂ90%) and that 27% of childhood precursor-B-ALL had TCRD gene rearrangements and/or deletions with germline TCRG and TCRB genes (Table 5).…”
