Immunomodulatory vitamin D effects on regulatory T-cells and cytokines in anin vitrostudy on patients with systemic lupus erythematosus

Abstract: Systemic lupus erythematosus (SLE) is a chronic autoimmune disease affecting various organs. Decreased numbers of regulatory T-cells (Treg cells; CD4 + CD25 high Foxp3 + ) are associated with the pathogenesis of SLE. A vitamin D deficiency was observed in many lupus patients. In the present study, peripheral blood mononuclear cells were isolated and cultured in the presence or absence of vitamin D, and total Tregs percentage was analyzed by flow cytometry. In addition, the level of expressions of Foxp3, TGFβ, … Show more

“…This also needs further clarification. However, consistent with this notion, Tregs have been shown to be dysfunctional in a state of vitamin D deficiency, and can have their functional status restored by sunshine, phototherapy and oral or topical vitamin D [ 153 , 154 , 155 , 156 , 157 , 158 , 159 , 160 , 161 , 162 , 163 , 164 , 165 , 166 , 167 , 168 , 169 , 170 , 171 , 172 , 173 , 174 , 175 , 176 , 177 , 178 , 179 , 180 , 181 , 182 , 183 , 184 , 185 , 186 , 187 , 188 , 189 , 190 , 191 , 192 , 193 , 194 , 195 , 196 , 197 , 198 , 199 , 200 , 201 , 202 , 203 , 20...…”

“…SLE is an autoimmune disease with abnormal innate and adaptive immune responses. TLRs are among the key components of innate immunity and their role in the activation of auto-reactive B lymphocytes and initiation of autoimmunity has been shown in various studies [Deng et al, 2013;Wang et al, 2014;Guo et al, 2015;Ghoryani et al, 2016;]. In SLE patients, endogenous products derived from dead cells and immune complexes activate TLRs signaling, which could in turn hyper-activate the immune responses [Ma et al, 2015].…”

“…It also blocks differentiation of monocytes into dendritic cells (DC), decreases the production of IL-12, INF-γ, IL-2, and enhances IL-10 production [ 6 ]. Other roles of VitD include increased monocyte-macrophage differentiation and control over macrophage responses [ 4 - 5 , 7 - 8 ].…”
