Immunomodulatory tetracyclines shape the intestinal inflammatory response inducing mucosal healing and resolution

Abstract: Immunomodulatory tetracyclines potentiate protective immune pathways leading to mucosal healing and resolution, representing a promising drug reposition strategy for the treatment of intestinal inflammation.

“…Indeed, it was demonstrated with in vitro and ex vivo studies how a short-term treatment with broad-spectrum antibiotics deeply affected both cellular and humoral immune response [ 79 , 80 , 81 ]. Some antibiotics have been reported to display immunomodulatory effect in addition to their antimicrobial activity [ 82 , 83 , 84 ]. Konstantinidis et al demonstrated that macrolides such as clarithromycin can induce Neutrophils Extracellular Trap (NET) generation both in vitro and in vivo.…”

“…Previous studies have shown that microRNAs are considered as significant regulators in the maintenance of epithelial barrier function [ 131 ], cell apoptosis [ 132 ], cell proliferation and differentiation [ 133 , 134 ], and immune functions [ 135 , 136 ]. It was found that microRNAs can trigger apoptosis and differentiation of intestinal epithelial cells and immune cells, which modulate the overall intestinal immunity for maintaining a delicately balanced status of intestinal environment [ 137 , 138 , 139 ]. In addition, it has also been reported that the microRNAs secreted by host intestinal cells can regulate the growth of microbes and the abundance of intestinal microflora via DNA exchanging, impact the target bacteria at the DNA level.…”

“…For example, macrolide antibiotics are used in the treatment of several chronic inflammatory diseases, such as chronic obstructive pulmonary disease (COPD) (Uzun et al, 2014), asthma (Gibson et al, 2017), and non-cystic fibrosis bronchiectasis (Li et al, 2019), due to their immunomodulatory properties. Minocycline and doxycycline have shown beneficial effects on experimental colitis (Garrido-Mesa et al, 2018).…”
