Leishmaniasis is an infectious disease that affects millions of people worldwide, making the search essential for more accessible treatments. The species Platonia insignis Mart. (Clusiaceae) has been extensively studied and has gained prominence for its pharmacological potential. The objective of this work was to evaluate the antileishmania activity, cytotoxic effect and activation patterns of macrophages of hydroalcoholic extract (EHPi), ethyl acetate fractions (FAcOEt) and morelloflavone/volkensiflavone mixture (MB) from P. insignis flowers. EHPi, FAcOEt and MB demonstrated concentration-dependent antileishmania activity, with inhibition of parasite growth in all analyzed concentrations. EHPi exhibited maximum effect at 800 μg/mL, while FAcOEt and MB reduced the growth of the parasite by 94.62% at 800 μg/mL. EHPi, FAcOEt and MB showed low cytotoxic effects for macrophages at 81.78, 159.67 and 134.28 μg/mL, respectively. EHPi (11.25 µg/mL), FAcOEt (11.25 and 22.5 µg/mL) and MB (22.5 µg/mL) characterized the increase in lysosomal activity, suggesting a possible modulating effect. These findings open for the application of flowers from a P. insignis flowers and biflavones mixture thereof in the promising treatment of leishmaniasis.

“…Estudos de toxicidade de doses repetidas aconselham a observação dos animais por, no mínimo, 2 semanas, caracterizando o perfil toxicológico da substância pela administração repetida, avaliando os quadros de: mortalidade, sinais clínicos (incluindo parâmetros comportamentais); variações no peso corporal e no consumo de ração e água, patologia clínica (hematologia, bioquímica); duração e reversibilidade da toxicidade . Desse modo, a baixa toxicidade do EH-Pi, no tratamento agudo, pôde ser comparado aos resultados também expressos por Lustosa et al (2016), em relação ao peso dos animais, para o extrato das sementes de P. insignis e por , através de estudos pré-clínicos em ratos, com um composto isolado da P. insignis, a Garcinialiptona FC (GFC), respectivamente. Objective: to identify the pharmacological activities of bacuri butter (Platonia insignis Mart.). Methods: an integrative review, carried out in the databases of Latin American and Caribbean Literature in Health Sciences, Cumulative Index to Nursing and Allied Health Literature, EMBASE, MEDLINE/PubMed, Web of Science, Cochrane Library and SCOPUS, without the time and language restriction. The selection consisted of 13 pre-clinical trials. The information assessment descriptively took place, comparing with the pertinent findings. Results: it was observed that 50.0% of the publications were indexed in MEDLINE/PubMed, most publications were from England (61.5%), followed by Brazil and the United States, both with 13.3%. It is noteworthy that 100.0% of the articles were pre-clinical trials; pharmacological activities for antioxidants (38.4%) and antileishmanicides (30.7%). It was found that 38.4% of the trials presented toxicity tests. Conclusion: bacuri butter (Platonia insignis Mart.) Showed pharmacological activities in pre-clinical trials, such as antioxidants, antileshimaniasis, anticonvulsant and wound healing.
