Background:The active components of natural products provide a potential alternative to conventional immunotherapy for a variety of diseases conditions and become subject to scientific investigations currently worldwide. Objective: The purpose of this research was to investigate the immunostimulatory activity of the chitosan nanoparticle on Wistar albino rats. Materials and Methods: The present investigation was carried out on various groups of healthy adult rats. The assessment of immunomodulatory potential was carried out by neutrophil adhesion test, delayed-type hypersensitivity (DTH) response, haemagglutinating antibody (HA) titre, cyclophosphamide-induced myelosuppression and phagocytic activity were determined in various groups of animals. Results: The administration of chitosan nanoparticle at doses 300 mg/kg BW and 600 mg/kg BW but not at doses 150 mg/kg BW significantly increased in neutrophil adhesion fibers, haemagglutinating antibody titre values and potentiated the inhibited type hypersensitivity reaction induced by sheep red blood cells. Also, it had good response towards phagocytosis in carbon clearance assay and prevented myelosuppression of cyclophosphamide on rats. Conclusion: From these findings, it can be concluded that chitosan nanoparticle responsible for immunostimulatory activity and has therapeutic potential for the prevention of immune depressed conditions.

Agaricus bisporus is one of the most consumed culinary mushrooms in the world. The white type of Agaricus bisporus has been demonstrated with various bioactivities beneficial to human health. However, little was known about its brown type. In this study, the anti-cancer and immunoregulatory potential of Agaricus bisporus (brown) polysaccharide were investigated in vitro and in vivo. In vitro, the polysaccharide had a significant antiproliferative effect on Hela cells (cervical cancer cells), especially at the high concentration of 200 μg/ml. The spleen index, thymus index and carbon clearance ability of mice were remarkably increased by the polysaccharide treatment, indicating the polysaccharide had a promising benefit for immunodulation in vivo. Furthermore, the polysaccharide increased the proliferation of RAW264.7 cells with 19.1% at 50µg /ml and 34.5% at 100µg /ml, which demonstrated the immunodulation mechanism was in close conjunction with macrophages. In conclusion, these results indicated that Agaricus bisporus (brown) polysaccharide possessed strong immunostimulatory and antitumor bioactivity in vivo and in vitro. Therefore, it could be explored as a novel natural antitumor agent with immunomodulatory activity.

Objectives: To scrutinise the immunostimulatory activity of methanolic extract of bark of Mimusops elengi Linn (MEMEL) in mice. Materials and Methods: The MEMEL was administered orally at the dose of 10, 20, 40 mg/kg/day body weight in mice. The immunostimulatory activities on specific and non-specific immunity were studied by carbon clearance test (CCT), haemagglutination antibody (HA) and delayed type hypersensitivity, using sheep red blood cells (SRBC) as the antigen. Distilled water served as a control in all the tests and vitamin E 150 mg/kg was used as standard. Result: Oral administration of MEMEL showed a dose-dependent increased immunostimulatory response. Phagocytic index was found to be increased significantly (P < 0.01) in CCT. The production of circulatory antibody titre (humoral antibody response) was increased significantly (P < 0.01) and delayed type hypersensitivity reaction was found to be augmented less significantly (P < 0.05) by increasing the mean footpad thickness at 48 hr in response to SRBC as an antigen. Conclusion: This finding suggests that the MEMEL possesses potential for augmenting immune activity by cellular and humoral-mediated mechanisms.
