Immunological studies on the respiratory burst oxidase of pig blood neutrophils

Abstract: Recently, a flavin enzyme (pI 5.0), that is probably responsible for superoxide (O~)-generating oxidase activity, was separated by isoelectric focusing-polyacrylamide gel electrophoresis (IEF-PAGE) from neutrophil membranes in our laboratory [(1987) J. Biol. Chem. 262, 12316-12322]. In the present work, we performed immunological studies on this enzyme derived from pig blood neutrophils. The enzyme extract obtained on IEF-PAGE was injected into guinea pigs to raise antibodies. IgG antibody against the pI 5.0 p… Show more

“…Earlier studies in neutrophils have noted an unidentified 18kDa polypeptide in the membrane fraction that was also the antigen recognized by NOX2 inhibiting antibodies [27, 28]. Further, a monoclonal antibody raised against TSPO was able to dose-dependently and specifically enhance NOX2 activity (i.e., the oxidative burst) in neutrophils [29].…”

“…This antibody was shown to dose-dependently inhibit the superoxide (02")generating activity of a membrane fraction isolated from leukocytes to approximately 50% of the total activity. Fukuhara et al (1988) studied a blood neutrophyl-derived flavin . enzyme involved in O2"-generating oxidase activity.…”
