Immunological Properties of Extraembryonic Human Mesenchymal Stromal Cells Derived from Gestational Tissue

Abstract: Mesenchymal stromal cells (MSCs) have been isolated from many tissues, including gestational tissue. To date, a study comparing the properties and suitability of these cells in cell-based therapies is lacking. In this study, we compared the phenotype, proliferation rate, migration, immunogenicity, and immunomodulatory capabilities of human MSCs derived from umbilical cord lining (CL-MSCs), umbilical cord blood (CB-MSCs), placenta (P-MSCs), and Wharton's jelly (WJ-MSCs). Differences were noted in differentiatio… Show more

“…Fetal MSCs can be isolated from different gestational tissues, such as umbilical cord lining and blood [41], Wharton's Jelly [42], amniotic fluid [4], and amnion and chorion layers [43]. Among them, c-kit pos AFS cells resulted as broadly multipotent SCs with intermediate features between embryonic SCs and MSCs, as they are capable of differentiating in vitro into cells of the three germ layers [4] and forming embryoid bodies [44].…”

“…The therapeutic properties of MSCs may vary substantially depending on their origin (1)(2)(3)(4)(5). Few data are available on clinical experience with MSCs from bone marrow (BM) or from other sources in similar patient groups.…”

“…decidua) and umbilical cord (UC) matrix (e.g. Wharton's jelly) are easily accessible without adverse donor site morbidity (2,3). Large quantities of potent "young" cells with high proliferative capacity can be obtained after short ex vivo expansion.…”

“…Another unique feature of CBMSCs is their low immunogenicity, which makes them suitable for allogenic cell therapies [14][15][16].…”
