SummaryThe present study was performed to investigate the clinical impact and certain virological and haematological parameters following immunization of cattle against lumpy skin disease (LSD). The study was conducted in a dairy cattle farm (215 animals), immunized with a Neethling strain-based live vaccine. Twenty-seven animals (14 lactating cows, four dry cows and nine calves) were randomly selected for repetitive blood and saliva samplings. An EvaGreen-based real-time PCR was designed to differentiate vaccine from field LSDVs. Vaccinated animals underwent examination for adverse reactions. Nodule samples were collected from two representative cases for histopathological testing and virus identification. Milk yield was calculated based on bulk-tank measurements of all lactating cows (79). Viral DNA was detected between days 6-15 post-vaccination (p.v.) at 63% of the sampled animals (17/27). Saliva and bulk-tank milk samples were LSDV-negative. Pronounced swelling was observed at injection sites of 12% of the immunized animals (26/215), starting at day 6 p.v., and was resolved after 2-4 days. Small-sized (<0.5 cm) cutaneous lumps were developed between days 8-18 p.v. at 9% of the vaccinated animals (19/215). These were observed in adult cows and not in calves/heifers.Resolution was observable 10 days post-development. The vaccine virus was also identified in nodules and injection-site aspirates. Haematological changes (e.g., lower leucocyte counts) were observed in cows and not in calves. Daily milk production was being reduced during the first 12 days p.v. LSD immunization of cows resulted in nodules and low viraemia levels. The fact that nodules and haematological changes were not observed in calves, along with the low viraemia, supports the reduced virulence of the Neethling vaccine strain. The characteristic nodules in vaccinated animals could allow clinical differentiation from those observed in LSD. The developed real-time PCR efficiently differentiates infected from vaccinated cattle, and should be further validated as a tool in LSD surveillance. † The first two authors contributed equally to this work.

Three hundred Oreochromis niloticus were divided into five groups; the control fish (CNT) were fed a basal diet, beta glucan (βG) group was fed 0.5 g/kg−1 βG, atrazine (ATZ) group was exposed to 1/5 96‐hr LC50 (1.39 mg/L) ATZ, the (βG/ATZ) group was fed βG while being exposed to ATZ, and the (βG then ATZ) group was supplemented with βG for fifteen days before exposed to ATZ. ATZ exposure caused a decline in growth that was ameliorated by βG. ATZ reduced the levels of total and different types of leucocytes. Additionally, ATZ exposure caused reductions in total proteins, globulins, α 1‐globulin, α 2‐globulin, ɤ‐globulin, immunoglobulin M, lysozymes, superoxide dismutase, nitric oxide and catalase but increases in hepatic transaminases and malondialdehyde without any variations in albumin and β‐globulin. Exposure to ATZ also resulted in a rise in the mRNA level of IL‐8. In contrast, expression of IgM, SOD and CAT were decreased in the tilapias exposed to ATZ. Exposure to ATZ increases the susceptibility response to Aeromonas sobria challenge, as indicated by an increase in cumulative mortality post‐challenge. Supplementation with βG fifteen days before (βG then ATZ group), counteracted the adverse effects of ATZ on the immune, biochemical and antioxidants values, though only slight alleviation was observed with simultaneous treatment (βG/ATZ group). Our results established that ATZ has adverse impacts on immune responses, antioxidant equilibrium and its related genes. While, supplementation with βG before exposure to ATZ may be valuable for counteracting the possible damage caused by ATZ water pollution than its simultaneous treatment with ATZ.

Lumpy skin disease (LSD) is an important disease of cattle. This study was conducted to determine alterations in hematological, biochemical and oxidative stress markers in cattle that have been naturally infected with LSD virus (LSDV). Blood samples and skin nodular lesions were collected from clinically infected, recovered and healthy animals. Quantitative real-time PCR was used to screen for Capripoxvirus DNA in samples from clinically infected animals. Hematological, biochemical and oxidative stress markers were measured. LSDV nuclic acids were detected in the collected samples using PCR. Hematological results revealed erythrocytosis, thrombocytopenia and leukopenia in infected cattle. Biochemical analyses showed that total protein and globulin levels were significantly elevated, while albumin and glucose were significantly reduced in these cattle. Aspartate aminotransferase (AST), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), creatine phosphokinase (CPK), and creatinine levels were markedly elevated. Moreover, serum levels of reduced glutathione (GSH) were markedly lowered, whereas lipid peroxidation (MDA) and inflammatory cytokines (IL-4 and TNF-α) were elevated. Recovered cattle exhibited significant amelioration of the alterations resulting from LSDV infection. The results of this study suggest that LSDV infection induces changes in hematological and biochemical parameters and stimulates oxidative stress; these findings may be helpful for choosing a good strategy for rapidly detecting and diagnosing LSDV infection.
