Immunological Aspects of AXL/GAS‐6 in the Context of Human Liver Regeneration

Abstract: AXL and its corresponding ligand growth arrest-specific 6 (GAS-6) are critically involved in hepatic immunomodulation and regenerative processes. Pleiotropic inhibitory effects on innate inflammatory responses might essentially involve the shift of macrophage phenotype from a pro-inflammatory M1 to an anti-inflammatory M2. We aimed to assess the relevance of the AXL/GAS-6-pathway in human liver regeneration and, consequently, its association with clinical outcome after hepatic resection. Soluble AXL (sAXL) and… Show more

“…In fact, HSCs express Gas 6 and Protein S, which are required as an insert to complete ABs internalization and the engulfment of TAMRA-labeled ABs, followed by profibrotic gene activation in LX2 cells, was significantly attenuated by Axl inhibitor. Given that Axl and Gas6 are notoriously involved in ABs engulfment by liver resident macrophages [ 72 , 73 , 74 ], our observation of Axl, Gas 6 and Protein S involvement in HSC engulfment of AB AGS+HIV confirms that this mechanism works for HSCs exposed to ABs.…”

“…70 In the same way, an increase in soluble AXL (sAXL) and GAS6 serum levels was observed after liver resection in cancer patients suggesting a strong contribution of GAS6-AXL signaling in liver regeneration. 71 Moreover, these changes were not observed in patients with high preoperative levels of sAXL and GAS6, which correlated with higher risk of liver dysfunction and worse clinical outcome.…”

“…During the regenerative process, Kupffer cells switch from the pro-inflammatory M1 to the anti-inflammatory, pro-regenerative M2 phenotype. Inhibited phenotype change in Kupffer cells has been shown to negatively impact postoperative liver regeneration and promote occurrence of PHLF 44 . See Fig.…”
