Immunoinformatics analysis and evaluation of recombinant chimeric triple antigen toxoid (r-HAB) against Staphylococcus aureus toxaemia in mouse model

Abstract: Staphylococcus aureus is a serious pathogen unleashing its virulence through several classes of exotoxins such as hemolysins and enterotoxins. In this study, we designed a novel multi-antigen subunit vaccine which can induce innate, humoral and cellular immune responses. Alpha hemolysin, enterotoxins A and B were selected as protective antigens for combining into a triple antigen chimeric protein (HAB). Immunoinformatics analysis predicted HAB protein as a suitable vaccine candidate for inducing b… Show more

“…The cytotoxic effects of SeNPs on both normal and cancerous cell populations were measured using the MTT assay, which was determined by the cellular proliferation rate and cell viability (Swaminathan et al, 2019(Swaminathan et al, , 2021Kota et al, 2021). The methodology is provided in the Supplementary Data [(10) Cell viability and micro-morphology assessment].…”

“…Because the OmpL of S. typhimurium which we considered for analysis is highly conserved among other Enterobacteriaceae pathogens in terms of its sequence and structure, this protein can be used as a broad-spectrum recombinant subunit vaccine for the gastrointestinal infections caused by the Enterobacteriaceae pathogens. Similar to our previous reports on subunit vaccine development, the OmpL can be cloned and expressed and can be administered to mice along with the potential adjuvants like Freund’s adjuvants to induce strong immunogenicity [ 8 , 9 , 14 ].…”

“…Liver models were assembled by staggered seeding of vascular and hepatic cell layers. In each sterilized biochip, 2.7x10 5 HUVECs/cm 2 and 1x10 5 monocytes/cm 2 were mixed and seeded on top of the membrane. HUVEC/monocytes were cocultured for five days with a daily medium exchange with vascular perfusion medium: endothelial cell medium supplemented with 20% autologous human serum, 10 ng/ml human granulocyte-macrophage colonystimulation factor (GM-CSF) (Peprotech), and 10 ng/ml human macrophage colony-stimulating factor (M-CSF) (Peprotech) to induce macrophage differentiation.…”

“…Infections with S. aureus are thus a leading cause of death in developed countries [3], with the MRSA strain USA300 reaching pandemic status across North America [4]. To date, no approved vaccine for S. aureus infections is available [5].…”
