Immunohistochemichal expression of biomarkers: a comparative study between diagnostic bronchial biopsies and surgical specimens of non-small-cell lung cancer

Abstract: Background:The increasing use of biomarkers as molecular determinants of responsiveness to conventional chemotherapy or molecular targeted therapy has raised the question of the reliability and reproducibility of their evaluation in bronchial biopsies as compared with corresponding resected surgical specimens. Results: High correlation coefficients were observed between the expression of ERCC1, hTERT and Ki-67 in the biopsies and the surgical specimens [0.83 (P < 0.0001); 0.55 (P < 0.001) and 0.64 (P < 0.0001)… Show more

“…It has been shown that ERCC1 immunodetection faces several methodological challenges including tissue processing, rates of interobserver agreement, geographic variation in protein expression and, most importantly, lack of the standardized cutoff value in semi-quantitative IHC [39][40][41]. It has been also reported that the most frequently used antibody 8F1 has not been specific for ERCC1 [42].…”

“…Cells (1x10 4 ) were seeded in 24-well plates with 18 duplications for each cell sample: H1299 cells (parental cells as control), H1299/E cells (transfected with PCDNA3.1-ERCC1) and H1299/V cells (transfected with PCDNA3.1 vehicle only). Survival cell of each cell sample were counted in three duplications every day.…”

“…Survival of the cells were evaluated under the inverted microscope (Olympus, Tokyo, Japan). Cell number was calculated according to the formula N=(n/4)x10 4 xV. Finally, cell growth curves were drawn with cell number as the y-axis and time as the x-axis.…”

“…In our study, 15% of the NSCLC patients had ERCC1 positivity. In a study of 34 NSCLC cases, a 7% higher ERCC1 expression was determined and a correlation between the ERCC1 expression and tumor cell type was reported [10]. In another study, ERCC1 expression was significantly higher in metastatic tissues compared with primary tumor tissues [11].…”
