“…However, cerebellar Purkinje cells expressing large amounts of mGluR1 mRNA as well as mGluR1a protein Baude et al, 1993;Görcs et al, 1993;Grandes et al, 1994) are not labeled by the present mGluR5 antiserum. Thus under the conditions of the immunocytochemical stainings, there is no evidence for such cross reactivity.…”

“…Both mGluR1 and mGluR5 are expressed in the cerebellar cortex with distinct cellular distribution patterns. Purkinje cells express high levels of mGluR1 mRNA , and immunohistochemical studies revealed that the mGluR1 protein is localized in spiny protrusions of the Purkinje cells postsynaptic to parallel fiber terminals (Baude et al, 1993;Görcs et al, 1993). This localization is consistent with the functional significance of mGluR1 for the induction of the long term depression (LTD) of the parallel fiber-Purkinje cell synaptic transmission (Conquet et al, 1994).…”

“…Previous studies have shown that every SOM-expressing neuron in the rat hippocampus coexpresses the "a" spliced form of the metabotropic glutamate receptor 1 (mGluR1a) with a 1:1 correlation (Martin et al, 1992;Baude et al, 1993;Görcs et al, 1993;Blasco-Ibáñez and Freund, 1995;Kerner et al, 1997). Fluorescence immunohistochemistry was thus performed for mGluR1a.…”

“…The sections were washed overnight in PB at 4°C, cryoprotected with 10, 20, and 30% sucrose in PB, frozen and thawed twice in liquid N 2 , washed several times in PB, and incubated with monoclonal mGluR1a antibodies, previously characterised (Kiss et al, 1996), either using the cell culture supernatants diluted 1:1-1:20 or the purified IgG at a concentration of 0.01-0.1 µg/ml. The immunohistochemical protocol was as described previously (Görcs et al, 1993;Grandes et al, 1994;Kiss et al, 1996); using 3,3,-diamino-benzidine (DAB) as chromogen (0,0125%, and 0,005% hydrogen peroxide; time of incubation 5-15 min, at room temperature). After the incubation, 3-3 sections (from each animal) were osmicated (1% OsO 4 in PB for 1 hr), dehydrated, and flat-embedded in Durcupan ACM (Fluka, Buchs, Switzerland).…”

“…In fact, mGluR1a transcripts (Masu et al, 1991;Shigemoto et al, 1992) and mGluR1a immunoreactivity (Martin et al, 1992;Baude et al, 1993;Görcs et al, 1993;Ryo et al, 1993;Grandes et al, 1994;Nusser et al, 1994;Takács et al, , 1997 are present at high density in cerebellum, the highest density being observed in Purkinje cells (PCs) and especially in their dendritic spines, but the receptor is also present in some interneurons of the molecular layer and in some Golgi neurons in the granular layer.…”
