Immunohistochemical study of nuclear factor-κB activity and interleukin-8 abundance in oesophageal adenocarcinoma; a useful strategy for monitoring these biomarkers

Abstract: IHC, using antibodies against the NLS of p65, may be useful in monitoring overall NF-kappaB activity in oesophageal tissues. As IHC is amenable to high-throughput screening (whereas traditional electrophoretic mobility shift assay methods are not), this may lead to the development of a better screening tool for early cancer risk.

“…In line with NF-kB and COX-2, expression of individual or combinations of proinflammatory cytokines IL-1b, IL-6, IL-8, and TNF-a is significantly increased in Barrett's esophagus and EAC as compared with squamous epithelium (82)(83)(84). IL-1b and IL-8 expression levels also correlate with the stage of EAC (79).…”

“…RELA codes for a number of cytokines and growth factors that in turn activate STAT3 resulting in a positive feed-forward loop [71]. NF-kB transcription is up-regulated along the sequence of BE to EAC with concurrent down-regulation of its negative regulator I-kB [77].…”

“…The transcription factor NF-κB has been found to be upregulated along the sequence from BO to OAC in tissue samples, along with one of its target molecules, IL-8 [35,36]. NF-κB can also be activated by the bile acid deoxycholic acid, a component of refluxate, in oesophageal cells, as well as low pH [37][38][39] IL-8 was upregulated in the proximal segment of BO in patients compared with the distal segment, along with other pro-inflammatory cytokines like IL-1β.…”
