Immunohistochemical Profile of MYC Protein in Pediatric Small Round Blue Cell Tumors

Abstract: Deregulation of MYC oncoprotein in cancers can result from multiple oncogenic mechanisms. Although MYC translocations define Burkitt lymphoma and MYC protein expression is a poor prognostic factor in undifferentiated neuroblastomas, the distribution of MYC protein (c-MYC) across other pediatric small round blue cell tumors (SRBCT) has not been well characterized. We undertook this study to assess MYC protein expression in a large cohort of pediatric lymphomas, sarcomas, and other SRBCT. Tissue microarrays cont… Show more

“…The proportions of positive staining cells were scored as follows: 0 (0%), 1 (< 25%), 2 (25–75%), and 3 (> 75%). The final scores of < 4 and ≥ 4 was deemed as low and high expression, respectively [ 24 , 25 ].…”

“…Our case illustrated diffuse and strong MYC immunoreactivity, suggesting a similar role of MYC in the pathogenesis of NUTM1-rearranged tumors from bone and the potential sensitivity to MYC-targeting therapeutic options. 20,21 Nevertheless, the diagnostic value of MYC stain in NUTM1-rearranged tumors is most likely to be limited by low specificity due to its role in various human cancers, including subsets of rhabdomyosarcomas and Ewing sarcomas, 22 and the majority of CIC-DUX4 sarcomas, 23 which can show morphologic overlap with NUTM1-rearranged tumors.…”

“…NUT carcinomas show frequent reactivity to epithelial markers (various cytokeratins 45 and rarely Claudin-4 13 ), SOX2 46 , and MYC 47 antibodies. However, SOX2 and MYC are not specific stains for NUT carcinoma [48][49][50][51] . Expression of the squamous markers p40 and p63 is variable and can be completely negative 52,53 .…”
