Immunohistochemical localization of neurocan and L1 in the formation of thalamocortical pathway of developing rats

Abstract: We used immunohistochemistry to examine possible molecular interactions between the subplate and growing thalamocortical axons in rat fetuses. In the cortical anlage of embryonic day 16 (E16), the subplate first appeared below the cortical plate. Among chondroitin sulfate proteoglycans, phosphacan was uniformly distributed throughout the cortical wall, whereas neurocan was localized only in the subplate at E16. Neural cell adhesion molecules, NCAM-H, TAG-1, and L1, were detected in the cortical anlage. Both co… Show more

“…Different from the CB + or CR + axons, L1 + axons also extended laterally into the LP/VP (Figures 4 and 9). L1 also is expressed by the growing thalamocortical axons (Fukuda et al, 1997), but these axons had not yet reached the ventral forebrain at the ages considered here. Thalamocortical L1 + axons in the posterior limb of the internal capsule are shown in Figure 5B.…”

“…Next, wild type and NCAM null mutant E13.5 and E15.5 mice (Cremer et al, 1994) were immunolabeled with antibodies to NCAM or PSA along with L1, which labels TC axons (Chung et al, 1991; Fukuda et al, 1997; Demyanenko et al, 2011b). L1-immunoreactive axons expressed transmembrane NCAM isoforms, as detected with a monoclonal antibody (mAb P61) that recognizes the cytoplasmic domain of both NCAM140 and NCAM180 (Figures 2A–A” and 3A–A”; Gennarini et al, 1984), but not NCAM120.…”
