Immunohistochemical expression of hypoxia-inducible factor-1α in stromal cells of vaginal tissue in post-menopausal women with pelvic organ prolapse

Abstract: Background & objectives:Pelvic organ prolapse (POP) is a common medical condition that affects adult women of different ages. The support of a normal pelvic floor is the result of complex interactions between ligaments, muscles, connective tissue and vaginal walls. Hypoxia and oxidative stress can reduce protein synthesis in the pelvic muscles that may contribute to muscular atrophy. Hypoxia-inducible factor-1α (HIF-1α) is a transcriptional activator which, expressed in response to hypoxia, activates a number … Show more

“…There is a paucity of data on the role of HIF-1α in the female urogenital tract, although production of HIF-1α has been detected in vaginal stromal cells [ 45 ] and in placental cytotrophoblasts [ 46 ]. Indeed, treatment with AKB-4924-induced robust stabilization in our vaginal epithelial cell HRE-reporter cell line indicating HIF-1α is produced by this cell type.…”

“…Nasal obstruction and hypoxia are both stimuli for the growth and development of rats throughout the process. Moreover, prolonged hypoxia can cause significant pathological changes in organs such as the skull, eyes, kidney, cardiovascular system, and gastrointestinal tract (Damman, Bloks, & Daha, 2015; Jakus, Jakus, Aračić, Stipić, & Vilović, 2017; Kim, Fong, Pilowsky, & Abbott, 2018; Klemm, Hurst, & Dias Blak, 2019; Wang et al., 2013; Yuan et al., 2018). However, at the very early stage, hypoxia is not as significant in remodeling the condyles of rats as mouth breathing.…”

“…Expression of Estrogen-⍺ receptor and VEGF-A measurements were calculated based on the number of epithelial cells expressing estrogen-⍺ receptor and VEGF-A in the vaginal wall tissue determined by calculating the mean number of vaginal wall cells that react positively to monoclonal antibodies seen under a microscope with 600x magnification in 10 fields of view using the Olympus DP12 Digital Camera (10).…”
