Immunohistochemical Detection of CD10 with Monoclonal Antibody 56C6 on Paraffin Sections

Abstract: A b s t r a c tWe tested a total of 174 paraffin-embedded hematolymphoid neoplasias to determine whether CD 10 can he specifically and sensitively detected on paraffin sections using monoclonal antibody 56C6 after epitope retrieval. For Material and Methods Case SelectionThe hematology and surgical pathology files of the Charite Hospital (Berlin, Germany) were surveyed to retrieve paraffin-embedded infiltrates of acute leukemias, malignant non-Hodgkin lymphomas, and myelomas from a total of 174 patients. In… Show more

“…Bcl-6 and CD10 are both markers of follicle center cells and are immunoreactive in noncutaneous follicle center lymphomas (16,40,41 (33). CD10 immunoreactivity is in keeping with Norton's (8) view that the true FCL of the skin have a similar immunophenotype to nodal FCL.…”

“…NEP was identified using a mouse monoclonal antibody to human NEP/CD10 (56C6, Novocastra Laboratories, Newcastle, UK) at a dilution of 1:50. This monoclonal antibody is highly efficient for immunostaining of NEP even in paraffin-enbedded tissue sections [18, 19]. ET-1 was identified using a rabbit polyclonal antibody to human ET-1 (Peninsula Laboratories, San Carlos, Calif., USA) at a dilution of 1:500.…”

“…Immunophenotypic methods have played an important role in the categorization of B-cell lymphomas into those of FCC or non-FCC origin because there are antigens that, even if not completely specific, characterize FCC as well as pre-and post-FCC. CD10 is expressed by precursor B cells as well as normal and many neoplastic FCC (7)(8)(9)(10)(11)(12)(13). Bcl-2 protein expression is another characteristic feature of neoplastic follicular centers, although there are many other normal and neoplastic bcl-2-positive lymphoid cells (14 -24).…”
