Immunogenicity of Recombinant Maltose-binding Protein (MBP)–Gonadotropin Releasing Hormone I (GnRH-I)

Abstract: A recombinant fusion protein, maltose-binding protein (MBP) -gonadotropin releasing hormone I (GnRH-I), was produced by cloning a gene fragment encoding a tandem repeated GnRH-I hexmer peptide (GnRH-I6) into the pMAL-c4x vector which was subsequently expressed in E. coli TB1. MBPGnRH-I6 was affinity purified. MBP-GnRH-I6 was verified by SDS-PAGE and Western blot and immunogenicity tested in boars. Injection of the recombinant fusion protein into boars yielded a high-titer antibody specific for GnRH-I. This was… Show more

“…Fang et al [10] used recombinant DNA technology to form maltose binding protein–gonadotropin releasing hormone I (MBP–GnRH-I6) vaccines, which had success in affecting the reproductive systems of pigs [11]. The objective of the present study was to evaluate the ultrastructure of gonadotrophin cells in MBP–GnRH-I6 immunized pigs.…”

“…MBP-GnRH-I6 was prepared and using recombinant DNA techniques as has been previously described [10]. Nine milligrams of MBP-GnRH-I6 or MBP was dissolved in 9 mL phosphate-buffered saline (PBS) and 9 mL of Al(OH) 3 adjuvant (Tianbang, Nanjing, China).…”

“…For the past 2 decades, GnRH-I sterilization vaccines have been produced by coupling GnRH-I to an antigenic carrier protein using chemical conjugation techniques. In an attempt to overcome this problem, a recombinant fusion protein was developed to serve as a sterilization vaccine in our laboratory (Fang et al, 2009(Fang et al, , 2010b. “…However, such methods generally create some reaction products that require extensive purification in order to provide a homogenous protein (Ulker et al, 2005). The recombinant fusion protein consists of six copies of GnRH-I in a linear sequence (GnRH-I6) and maltosebinding protein (MBP) (MBP-GnRH-I6) (Fang et al, 2009(Fang et al, , 2010b.
