Salmonella enterica can be considered as one of the most important causes of foodpoisoning with poultry thought to be the main source. Although S. Typhimurium, S. Enteritidis and the vast majority of other Salmonella serovars generally produce little systemic disease in adult chickens, they are able to colonize the alimentary tract of poultry. The two caeca are the main sites of the colonization of Salmonellae in chickens, and the bacteria can be easily harvested from the caeca for analysis. Bacterial proteins analyses utilizing SDS-PAGE showed differences between in vitro and in vivo that out of about 40 protein bands of in vitro preparation only a few (3-5) bands can be visualized from in vivo preparations. We suggested that some avian proteases might be responsible. Accordingly, and to investigate the hypothesis that an inactivated vaccine harvested from chickens is thought to be more protective than bacteria grown in broth culture, the immunogenicity of inactivated vaccines, either proteins or formalin killed bacteria (S. Enteritidis) harvested from chicken intestine and those from broth culture (in vitro), were compared using bacterial proteins or formalin killed bacteria as an orally inoculated vaccine candidate in chicken. The results demonstrated that sonicated proteins extracts in general have a stronger effect as vaccines against S. Enteritidis colonization in chickens than formalin killed bacteria. In contrast, the in vitro sonicated proteins obtained from a nutrient broth culture had a much better protective vaccine effect than the in vivo sonicated proteins preparations harvested from bacteria grown in chickens. IntroductionMany bacterial pathogens such as Clostridium, Staphylococcus, Campylobacter and many other bacterial strains are capable of causing food-poisoning, and Salmonella enterica can be considered as one of the most important causes with poultry thought to be the main source. Although S. Typhimurium, S. Enteritidis and the vast majority of other Salmonella serovars generally produce little systemic disease in adult chickens, they are able to colonize the alimentary tract of poultry, resulting in contamination of poultry carcasses and entry into the human food chain. However, there is a great demand to control food-poisoning salmonellosis at both breeder and layer levels at the national and global level in order to produce Salmonelle-free poultry products, due to the current correlation between S. Enteritidis PT4 and poultry products. Salmonellosis costs the European Union a minimum of 500-900 million Euros annually. Salmonellosis in food animals is a major target for reduction of human infection by the European Union. Legislation has been introduced to monitor the most important Salmonellae serovars. ABSTRACT. Bovine bacterial respiratory diseases have been one of the most serious problems due to their high mortality and economic loss in calves. The vaccinations of bovine bacterial respiratory vaccines have been complex because of no multivalent vaccine. In this study, novel multivalent bovine bacterial respiratory vaccine (BRV) was developed and tested for its safety and efficacy. BRV was composed of two immunogens and five bacterins. These were leukotoxoid and bacterin of Mannheimia haemolytica type A, outer membrane protein and bacterin of Pasteurella multocida type A, and bacterins of Haemophilus somnus, Mycoplasma bovis, and Arcanobacterium pyogenes. ELISA antibody titers to five bacterial antigens in vaccinated guinea pigs increased, compared with those in unvaccinated ones. BRV was safe for calves and pregnant cattle in this study. In calves challenged with M. haemolytica and P. multocida, the average daily weight gain and antibody titers of vaccinated calves increased, and respiratory symptoms (P<0.05) and treatment frequency (P<0.01) of vaccinated calves significantly decreased, compared with those of unvaccinated calves. Interestingly, the antibody titers of M. haemolytica leukotoxoid and Mycoplasma bovis were closely related with the reduction of respiratory symptoms. BRV would be an ecomonical measure for the protection against bovine bacterial respiratory diseases. Serotype B of Pasteurella multocida is a major agent of haemorrhagic septicaemia (HS), a form of blood poisoning widespread in cattle and buffalo from Southern Asia. The vaccines currently used against the disease are not potent enough, being effective only for a limited duration only. In an attempt to identify immunogenic components of P. multocida for use as live/subunit vaccines, the outer membrane proteins (OMPs) of B:2 and B:3,4 serotypes were isolated and their profiles compared. Three common immunogenic components; polypeptides of molecular weight (MW) 39.0, 33.5, 31.0 kDa, were identified by immunoblotting. The immunogenicity of identified OMPs was also confirmed using antisera generated against them. The antigenic OMPs identified in current research could be explored in the formulation of new, safe and effective vaccines for indigenous buffalo and cattle breeds with a prolonged efficacy. Canlı/subunit aşılarda kullanılmak üzere Pasteurella multocida'nın immunojenik komponentlerinin belirlenmesi için yapılan çalışmamızda, B:2 ve B:3,4 serotiplerinin dış membran proteinleri (OMP) izole edilmiş ve bunların profilleri karşılaştırılmıştır. İmmunbloting sonucunda 39.0, 33.5 ve 31.0 kDa moleküler ağırlıklarında 3 adet ortak immunojenik polipeptit belirlenmiştir. İdentifiye edilen dış membran proteinlerinin immunojenitesi ayrıca bunlardan elde edilen antiserumlar ile doğrulanmıştır. Bu çalışmada identifiye edilen antijenik OMP'ler uzun süre etki gösteren yeni, güvenli ve etkin aşıların formülasyonlarının geliştirilmesine katkı sağlayacaktır.
