Immunogenic properties of empty pcDNA3 plasmid against zoonotic cutaneous leishmaniasis in mice

Abstract: BackgroundLeishmania(L) parasite, the causative agent of zoonotic cutaneous leishmaniasis (ZCL), effectively stimulates the mammalian cells to mount strong humoral responses by enhancing T-helper-2 (Th2)-associated cytokines for its survival. The best strategy to decrease the intensity of infection in the host is induction of cellular immunity.MethodsWe evaluated the effects of the empty bacterial pcDNA3 plasmid on mice infected withL.majorand quantified the immune mediators including IFN-γ, IL-4, IL-10, IgG2a… Show more

“…All of the split TEV assays were based on transient transfection with pcDNA3-based vectors, which are some of the most frequently used vectors in molecular and cellular biology [35]. However, we cannot rule out any differences in assay performance that may occur when using different vector backbones or different promoters (we used the CMV promoter; other options are, e.g., human EF1α or UbC promoters) for expressing split TEV fusion proteins.…”
