Immunoelectronmicroscopic examination of early lesions in histamine induced immune complex vasculitis in man

Abstract: In 4 cases of allergic vasculitis circulating immune complexes (IC) were demonstrated. Spontaneous and histamine induced vascular changes were studied by immunofluorescence microscopy. The early events in IC vasculitis were investigated at the ultrastructural level by immunoelectronmicroscopy using the peroxidase-antiperoxidase multistep technique. Our findings support the concept that human IC vasculitis is triggered by the deposition of circulating IC in the walls of postcapillary venules between endothelial… Show more

“…So we regard these findings to be an early phase of im mune complex vasculitis. This interpreta tion is in accordance with previous reports of direct IF on histamine-induced early vas culitis lesions in patients with allergic vascu litis [2,24], Interestingly, the BD patient had no oral aphthae at the time of investigation, the other patient had only C3 deposits in subepithelial vessel walls of oral aphthous lesion.…”

“…Though not revealed by direct IF in the present study, C3 deposits in lesional vessel walls may indicate the presence of immune complexes which already have been removed by chcmotactically attracted polymorphonuclear leukocytes. Detection of C3 alone in biopsies taken from visible vasculitic skin lesions is a common finding [2,22,24], Thus, tissue-bound C3 in late phase leukocytoclastic vasculitis only repre sents the remnant of the process leading to complement activation by immune com plexes.…”

“…We injected histamine in healthy skin of patients' legs to provoke circulating im mune-complex precipitation in dermal ves sel walls. This method has been widely used in a variety of dermatoses where immune complexes may play a role [7,8].…”
