Immunodominant antigen of Actinobacillus actinomycetemcomitans Y4 in high-responder patients

Abstract: This study was undertaken to look for characteristics of the immunodominant antigen(s) of Actinobacillus actinomycetemcomitans Y4 that might help explain the high antibody titers in periodontitis patients. Radioimmunoassays (RIA) were performed on sera from 481 patients; sera from the 32 patients with the highest anti-Y4 titers (above 128,000 RIA U/mI) were further analyzed. Y4 antigen was boiled for 45 min or treated with papain, and antibody responses were analyzed by RIA and Western blotting (immunoblotting… Show more

“…The specific strain of bacteria used may account for other differences in the antibody response in these "refractory" patients when compared to published data on antibody responses in other patient populations. For example, serotype antigens on A. antinomycetemcomitans are very immunogenic in humans (Califano et al 1989, Taichman et al 1982. Consequently, possible differences in the serotype of the plaque bacterium being tested compared to the antigens source used in the ELISA may have caused a decrease in the detection of total antibody against a specific periodontal pathogen.…”

“…These findings have also been confirmed by the routine detection of significant elevations in serum antibody coincident with the infection (Listgarten et al 1981, Ebersole et al 1982, Genco et al 1985, Tew et al 1985b, Gunsolley et al 1988, Ebersole 1990, Mooney et al 1993, Ebersole & Taubman 1994. More detailed investigations of this antibody response have delineated some antigenic specificity for various membrane proteins (Califano et al 1989, Watanabe et al 1989, Bolstad et al 1990, Sims et al 1991, Wilson 1991a, b, Saito et al 1993, Ebersole et al 1995a, Nakano et al 1995, carbohydrates (Califano et al 1989), and lipopolysaccharide epitopes , Wilson & Schifferle 1991, Wilson & Hamilton 1992. The majority of these reports described cross-sectional analyses of antibody responses in rather heterogeneous populations of periodontitis patients.…”

“…Various investigators have demonstrated that patients with localized juvenile periodontitis exhibit elevated serum IgG antibody to A. actinomycetemcomitans and that the immunodominant antigen of the organism is a capsular-like polysaccharide or the polysaccharide portion of lipopolysaccharide (6,10,23,26,36). In addition, serum IgG antibody to A. actinomycetemcomitans elevated in periodontitis patients has been reported to enhance in vitro phagocytosis of the organism by human neutrophils (3,27,34).…”

“…The serotype-specific antigens of serotypes d and e have not yet characterized. Patients with localized juvenile periodontitis exhibit elevated serum antibody levels to SPA of A. actinomycetemcomitans serotype b (6,23,26,36). However, the role of serum SPA-specific antibodies in host responses to A. actinomycetemcomitans is poorly understood.…”
