Immunocytochemical localization of neutrophil elastase in term placenta decidua and myometrium in pregnancy‐induced hypertension

Abstract: Objective-The aim of the study was to determine whether there was evidence of elastase containing neutrophils at the materno-fetal interface in women with pregnancy-induced hypertension (PIH). Design-An observational prospective study. Setting-The Simpson Memorial Maternal Pavilion Edinburgh.Merhods-Placentas were obtained at vaginal or abdominal delivery from 5 1

“…These results coincide with previous studies demonstrating that choriodecidual leukocytes express CAMs [16,39,40], cytokines [8,41,42], chemokines [16,34], and their receptors [16,34,43], enzymes [35,44,45], antimicrobial peptides [46], markers of activation [17,39,[47][48][49], and even hormone receptors [50] before and during the process of labor. Together, these findings demonstrate that the choriodecidual leukocytes are a rich source of inflammatory mediators that can participate in the proinflammatory milieu that accompanies the process of labor.…”

“…In fact, the findings suggest an enhanced capacity for all the steps in leukocyte/endothelial interaction, including capture, rolling, firm adhesion, and transmigration in preeclampsia. This may be the mechanism behind the increased infiltrate of neutrophils 19 and mononuclear perivascular cells 20 found in the decidua in preeclampsia.…”

“…2 Immune abnormality has been speculated to lead to cellular activation and release of substances that alter maternal endothelial function in a direction that causes vascular leakage, coagulopathy, and hypertension. Evidence suggests that neutrophils are activated in both the placental bed 3 and maternal circulation of women with preeclampsia 4 -8 and that this activation resolves after delivery. 9 Neutrophil activation is associated with free radical release that either can affect endothelial function directly or contribute indirectly through production of lipid peroxides.…”
