A single injection of estradiol benzoate (10 pg), while highly effective in ovariectomized female guinea-pigs, does not prime castrated males to display progesterone-facilitated lordosis. In contrast, adult males and females exhibit the same, high degree of progesterone-facilitated lordosis when primed with two, 2.0 pg injections of free estradiol-17B (pulse regimen). We compared neural progestin receptor induction after these different estradiol treatments by in vitro radioligand binding assays and imrnunocytochemistry. Binding assays confirmed previous observations of lower concentrations of cytosol progestin receptors in the rnediobasal hypothalamus-preoptic area in estradiol benzoate-treated males than in females. No such sex difference was observed in animals that had been exposed to the behaviorally effective estradiol pulse regimen; rather, hypothalamic-preoptic area progestin receptor concentrations in these animals did not differ from the low levels observed in males treated with the behaviorally ineffective estradiol benzoate regimen. lmmunocytochemical analysis revealed progestin receptor-immunoreactivity in fewer cells in the medial preoptic nucleus-anterior hypothalamic nucleus, periventricular preoptic area and arcuate nucleus of estradiol pulse-as compared to estradiol benzoate-treated males and females. Estradiol benzoate treatment induced progestin receptor-immunoreactivity in more cells in the medial preoptic area and ventrolateral hypothalamus than estradiol pulses in males, but not in females. Surprisingly, in these regions estradiol benzoate-treated males had significantly more progestin receptor-immunoreactive cells than females.These experiments yield two major findings: First, as has been shown in rats, the display of progesterone-facilitated lordosis is not inflexibly differentiated according to sex in guinea-pigs. Furthermore, reduced concentrations of estradiol-induced progestin receptors in the hypothalamus and preoptic area cannot account for the lack of progesterone-facilitated lordosis that is observed following priming with estradiol benzoate in males. Secondly, in the medial preoptic area and ventrolateral hypothalamus of female guinea-pigs, estradiol pulses are as effective as estradiol benzoate in inducing progestin receptors. These observations, taken together with the finding of equal behavioral efficacy of the two estradiol treatments, are consistent with the hypothesis that estradiol-induced progestin receptors in these regions of the brain are involved in progesterone-facilitated lordosis in female guinea-pigs.The display of feminine sexual behavior in many rodents, including guinea-pigs, is believed to be sexually differentiated. Progesterone receptor immunoreactivity in aromatic L-amino acid decarboxylase-containing neurons of the guinea pig hypothalamus and preoptic area

A double-labeling immunofluorescence procedure was used to determine whether progesterone receptor (PR)-immunoreactive (IR) neurons in the preoptic area and hypothalamus of female guinea pigs also contained aromatic L-amino acid decarboxylase (AADC), an enzyme involved in the synthesis of both catecholamines and serotonin. Immunostaining was performed on cryostat sections prepared from ovariectomized guinea pigs primed by estradiol to induce PR. The nuclear presence of PR was visualized by a red fluorescence while the AADC-containing perikarya showed a yellow-green fluorescence. The topographic distribution of AADC-IR neurons was investigated by using a specific antiserum obtained by immunization of rabbits with a recombinant protein beta-galactosidase-AADC in the two regions known to contain the densest populations of estradiol-induced PR-IR cells: the preoptic area and the mediobasal hypothalamus. The localization of PR-IR and AADC-IR cell populations showed considerable overlap in these areas, mainly in the medial and periventricular preoptic nuclei and in the arcuate nucleus. A quantitative analysis of double-labeled cells estimated that about 15% to 23% of AADC-IR cells in the preoptic area and about 11% to 21% of AADC-IR cells in the arcuate nucleus possessed PR. This colocalization persisted throughout the rostrocaudal extent of these areas and represented 3% to 9% of the population of PR-IR cells. These findings provide neuroanatomical evidence that a subset of AADC neurons is directly regulated by progesterone. The exact physiological role of this enzyme in target cells for progesterone is not understood. AADC may be involved in functions other than that for the synthesis of the classical neurotransmitters.

Ovarian hormones influence the morphology, distribution, and density of tyrosine hydroxylase immunoreactive axons in the dorsolateral prefrontal cortex of adult Rhesus monkeys

The maturation, adult functioning and dysfunction of the prefrontal cortex in disorders such as schizophrenia show gender biases in human and non-human primates. Although the basis for the hormone influence suggested in these observations is unknown, one possibility is that circulating hormones stimulate catecholamine innervation in the frontal lobe. This innervation is essential for prefrontal cortical function, and gonadal, especially ovarian hormones, profoundly influence catecholamine function and physiology in subcortical structures. This study was undertaken to determine whether influence is also exerted upon the catecholamine innervation of the association cortex by combining ovarian hormone manipulation with immunocytochemistry for tyrosine hydroxylase in the dorsolateral prefrontal cortex of adult female macaque monkeys. Qualitative and quantitative analyses of immunoreactive fibers were carried out and compared in cortices of ovariectomized animals, ovariectomized animals treated with estrogen, ovariectomized animals treated with estrogen followed by progesterone, and in intact, age- and sex-matched controls. These analyses revealed striking, layer-specific anomalies in fiber morphology and profound reductions in fiber density in ovariectomized animals. While hormone replacement with estrogen alone had limited influence, estrogen followed by progesterone was particularly effective in restoring tyrosine hydroxylase innervation in ovariectomized animals. Thus, ovarian hormones appear to be potent regulators of the catecholamine innervation of the primate prefrontal cortex. Such regulation is anticipated in the gender differences observed in prefrontal cortical development and function, and may also be relevant for the prefrontal dysfunction in disorders such as schizophrenia.
