Immunocytochemical demonstration of ecto-galactosyltransferase in absorptive intestinal cells.

Abstract: Galactosyltransferase immunoreactive sites were localized in human duodenal enterocytes by the protein A-gold technique on thin sections from low temperature Lowicryl K4M embedded biopsy specimens. Antigenic sites detected with affinity-purified, monospecific antibodies were found at the plasma membrane of absorptive enterocytes with the most intense labeling appearing along the brush border membrane. The lateral plasma membrane exhibited a lower degree of labeling at the level of the junctional complexes but … Show more

“…This is confirmed by the presence ofboth galactosyl transferase and Ig in this structure (Fig. 7 b), since this enzyme was never found in endocytotic vesicles but only in RER and Golgi complex (15,16). There is no background labeling, as can be seen over the mitochondria and nucleus (Fig.…”

“…Monospecific antibodies were obtained by repeated absorption on blots and release by exposure to glycine/HCl buffer, pH 2.8 (Fisher & Smith, 1988;Roth et al, 1985). After 1 min the fixative was diluted 1:10 for 14 min.…”

“…Excised and destained spots were impregnated with complete Freund's adjuvant and implanted under sterile conditions close to inguinal lymph nodes in adult male CHbb: CH rabbits. Sera were prepared from blood samples collected after 30 days.Monospecific antibodies were obtained by repeated absorption on blots and release by exposure to glycine/HCl buffer, pH 2.8 (Fisher & Smith, 1988;Roth et al, 1985). “…Galactosyltransferase has also been localized to the plasma membrane of a diverse variety of cells and tissues by immunohistochemical (5)(6)(7)(8)(9) and biochemical procedures [comprehensively reviewed in Pierce et al (10) and Shur (11)]. This cell surface distribution has led to the postulate that, in addition to its biosynthetic role, galactosyltransferase has a functional role in intercellular recognition and/or adhesion (12).…”
